消油剂对海湾扇贝(Argopecten irradians)的急性毒性效应   总被引:1,自引:0,他引:1       下载免费PDF全文
本研究以海湾扇贝(Argopecten irradians)为研究对象, 通过96 h半静水毒性实验,探讨了消油剂、0号柴油分散液(WAF)与0号柴油乳化液(CEWAF)对海湾扇贝24 h、48 h、72 h、96 h急性毒性效应。结果显示,消油剂对海湾扇贝的24 h、48 h、72 h和96 h半致死浓度(95%可信限)分别为1905(1570-2451 mg/L)、1293(1070-1556 mg/L)、1015(721-1348 mg/L)、846 mg/L(660-1020 mg/L);WAF对海湾扇贝的24 h、48 h、72 h和96 h半致死浓度(95%可信限)分别为36.57(29.42-48.96 mg/L)、23.10(21.07-25.39 mg/L)、14.99(13.66-16.41 mg/L)、11.31 mg/L(8.37-14.47 mg/L);CEWAF对海湾扇贝的24 h、48 h、72 h和96 h半致死浓度(95%可信限)分别为12.71(10.78-15.55 mg/L)、8.51 (7.99-9.07 mg/L)、6.56(4.46-7.74 mg/L)、5.42 mg/L(4.51-6.29 mg/L)。消油剂、0号柴油分散液、0号柴油乳化液对海湾扇贝96 h安全浓度分别为84.6、1.13、0.54 mg/L。随着实验时间的延长,消油剂、WAF和CEWAF对海湾扇贝的半致死浓度(LC50)均呈逐渐降低之势,半致死浓度与时间呈幂函数关系,其关系式分别为y=12242x–0.583、y=564.37x–0.848、y=89.987x–0.613。在受试溶液中暴露相同时间,海湾扇贝的半致死浓度从高到低依次为消油剂、WAF和CEWAF,说明CEWAF对海湾扇贝的毒性大于消油剂和WAF,这对于在养殖海区使用消油剂处理溢油事故具有重要的指导意义。  相似文献

ABSTRACT:   This study examines the toxicity of three dispersants and heavy fuel oil to a marine fish, red sea bream ( Pagrus major ). Also studied was the toxicity of a mixture of heavy fuel oil and the least toxic of the three dispersants. The 24-h LC50 of all three dispersants were at least 1500 mg/L; these dispersants appeared relatively less toxic to marine fish than others studied in the past. The mean lethal oil concentration of the water-accommodated oil fraction was 325 µg/L. Mixtures of oil and dispersant were more toxic than dispersant or oil alone. Large amounts of dispersant decreased the toxicity of the mixture for the marine fish. Use of a dispersant-to-oil percentage of 20%, which is recommended by the manufacturer because of its efficiency in oil emulsification and dispersion, yielded higher 24-h oil concentrations and resulted in a higher mortality rate than did the use of higher percentages of dispersant. The application of dispersant to oil in coastal areas, especially with higher activities of fisheries and aquaculture like Japan, must be considered carefully in the context of the benefits versus environmental cost.  相似文献

Rainbow trout,Oncorhynchus mykiss, were exercise-trained for 18 hours per day over 28 days at water velocities up to 60% of their measured Ucrit. Anin situ perfused heart preparation was used to compare maximum cardiac performance between control and trained fish. Trained fish had a larger stroke volume at a given filling pressure, as well as an 18% higher cardiac output and a 25% greater maximum power output. These observations indicate that exercise training in rainbow trout improved maximum cardiac performance. Adrenaline produced positive inotropic and chronotropic effects on the perfused heart, but exercise training did not alter these stimulatory effects. Maximal activities of citrate synthase (CS), B-hydroxyacyl CoA dehydrogenase (HOAD), glutamate dehydrogenase (GDH) and carnitine palmitoyl transferase (CPT) were measured in cardiac and skeletal muscles. CS, HOAD and GDH increased in red and white skeletal muscle as a result of training. Training also increased GDH activity in the endocardium and epicardium, and increased HOAD in the epicardium. While the training regime did not result in a statistically significant increase in Ucrit and produced a decrease in the condition factor of the fish, other training effects were clearly evident. Furthermore, significant correlations were observed between Ucrit and the maximal activities of GDH and HOAD.  相似文献

A 60-day feeding trial was conducted to determine the effects of palm oil blended with oxidized and non-oxidized fish oil on growth performances, hematology, and non-specific immune response in juvenile Japanese sea bass, Lateolabrax japonicas. Japanese sea bass (1.73 ± 0.01 g) were fed seven experimental diets containing 100 g/kg of dietary lipid in forms of palm oil (10P), fish oil (10F), fish oil blended with palm oil at different ratios, 6:4 (6F4P) and 4:6 (4F6P), oxidized fish oil (10OF), and oxidized fish oil blended with palm oil at different ratios, 6:4 (6OF4P) and 4:6 (4OF6P). After the feeding trial, the following results were illustrated. No significant effects were observed in survival, feed conversion ratio, condition factor, and hematocrit after feeding with experimental diets for 60 days. The relatively higher specific growth rate and hematology were observed in 6F4P. Furthermore, both palm oil and oxidized fish oil acted as a negatively on serum lysozyme activity (P < 0.05). This study suggested that a ration of 6F4P is recommended as an innocuous ratio for Japanese sea bass. Furthermore, according to the present investigation, palm oil seems to have the ability to improve the protein efficiency when added to oxidized fish diets as well as a positive trend to the growth performance (P > 0.05).  相似文献

In vitro methods have previously been utilised for the rapid and reliable evaluation of protein digestibility in fish. In this study we used in vitro methods to compare the digestibility of various live and artificial larval feeds and feed ingredients. Given previous suggestions that water-soluble dietary proteins are efficiently digested and utilised by stomachless fish larvae, we also analysed the content of water-soluble nitrogen in the feeds and feed ingredients and then measured the specific in vitro digestibility (simulated midgut conditions) of the water-soluble and insoluble fractions. The soluble nitrogen fractions were generally more digestible than the insoluble nitrogen fractions (P < 0.05). A soluble reference protein (Na+-caseinate) was digested faster than the similar but insoluble reference protein (casein) although their final digestibility was the same (94%). Frozen live feeds (Artemia fransiscana and Calanus finmarchicus) contained high fractions of soluble nitrogen (54–67%) and also had high digestibility in vitro (84 and 87%, respectively). The in vitro digestibility of two formulated larval feeds tested was lower (53 and 70%) than the frozen live feeds. The digestibility of the ingredients of the protein-encapsulated feeds particles was reduced as a result of the production process (from 71 to 53%, respectively). Three meals of marine origin (fish meal, squid meal and fish roe meal) all had low contents of water-soluble nitrogen (11–17%) but showed different degrees of digestibility (77, 77 and 49%, respectively). The results also demonstrated that while pre-hydrolysis of a feed ingredient (fresh frozen cod fillet) almost doubled the water-soluble nitrogen fraction (from 34 to 65%) the positive effect of pre-hydrolysis on in vitro digestibility was much lower (from 80 to 86%). This demonstrates the complexity of assessment of bioavailability of dietary protein sources in larval fish; a number of factors such as leaching rates from feed particles, digestibility, digestion rates and absorption rates all need to be taken into account.  相似文献

Inclusion of the water-soluble fraction of blue mussels Mytilus galloprovincialis as a feed additive was examined with tiger puffer Takifugu rupbripes. The control diet mainly consisted of fish meal, potato starch, and pollack liver oil. Experimental diets were formulated to replace 30% and 40% of the fish meal protein with defatted soybean meal (SBM), and were supplemented with 0–20% mussel extracts. Fish of 18 g initial body weight were fed each diet to satiation, twice daily, 6 days per week for 7 weeks at 20°C. Weight gain and protein efficiency ratio of fish fed the diet containing SBM without the extract were significantly lower than those of the control at the 30% replacement level. Addition of 5% or 10% extract improved the growth and feed utilization to levels comparable to those of the control. At 40% substitution, statistically identical growth and feed performance to the control were obtained for diets supplemented with 10% or 20% extract; however, performance for 5% addition was significantly lower. Thus, the water-soluble fraction of blue mussels would be an effective feeding stimulant in aquaculture feed for tiger puffer that makes it possible to replace 30–40% of fish meal protein with SBM.  相似文献

This study was carried out to investigate the effect of feeding diets containing herring or anchvoy oil, on flesh quality parameters of Atlantic salmon (Salmo salar). Two extruded experimental diets with the same basal composition but one coated with herring oil and the other with anchovy oil, were each fed during 24 weeks to salmon with an average initial weight of 1.8 kg. Salmon grew to a final weight of 3.9 kg. Growth, condition factor and biometric parameters were not affected by the dietary treatment. No significant differences were found for intramuscular fat. Monounsaturated fatty acid (MUFA) concentrations were highest in the group fed the diet containing herring oil, in both neutral and polar lipids, while the group fed the diet containing the anchovy oil showed a higher concentration of n-3 fatty acids in both fractions of intramuscular lipids. The n-3/n-6 ratio was higher in the neutral lipid fraction of fish fed the southern hemisphere oil, while no significantly differences were found for the polar lipid fraction. No differences were found on muscle α-tocopherol levels. Muscle homogenates from fish fed the anchovy oil showed the highest thiobarbituric acid reactive substances (TBARS) after 9 days of storage. However no differences were found between groups on the induced oxidation tests. It is concluded that the origin of the fish oil has no effect on growth perfomance, but there is a marked effect on fatty acid composition and susceptibility to lipid oxidation.  相似文献

为深入了解心型脂肪酸结合蛋白(heart-type fatty acid-binding protein,H-FABP)的结构与功能,采用RACE技术克隆得到暗纹东方鲀H-FABP的全长cDNA序列,共772 bp,开放阅读框为402 bp,编码133个氨基酸。生物信息学分析表明,暗纹东方鲀H-FABP基因具有一个脂质转运蛋白家族保守结构域和多个蛋白激酶磷酸化位点。NJ法系统进化分析显示,暗纹东方鲀与红鳍东方鲀亲缘关系最近,其次是鲈形目。经荧光定量PCR检测,H-FABP基因在暗纹东方鲀所有被检测的组织中都有表达,其中肝脏中的表达量最高,其次为肌肉和心脏,预示H-FABP主要参与脂肪酸氧化分解过程。为探究豆油作为脂肪源的可行性,采用不同比例(0、25%、50%和75%)豆油替代鱼油的饲料投喂初始体质量为(8.31±0.20)g的暗纹东方鲀8周。结果显示,全鱼油组肝脏中H-FABP基因的表达量显著高于50%豆油组和75%豆油组,但与25%豆油组无显著性差异,且随着饲料中豆油比例的增加,暗纹东方鲀肝脏中H-FABP基因的表达量呈下降趋势。研究表明,富含n-3多不饱和脂肪酸的鱼油可以促进脂肪酸氧化分解,能预防脂肪肝。因此,为不影响暗纹东方鲀肝脏脂肪代谢,饲料中豆油替代鱼油的比例应控制在50%以下。  相似文献

The aim of the study was to investigate the effects of the diets that contain safflower oil and fish oil as lipid sources, on growth, feed conversion and body composition in turbot (Psetta maxima). Two iso-nitrogenous and iso-lipidic diets (55 % protein and 14 % lipid) were prepared that include 100 % fish oil (FO group) and 100 % safflower oil (SFO group) for turbots with average weight of 62.21 ± 1.28 g, and fish were fed with these diets for 104 days. At the end of the experiment, the weight gain, specific growth rate and feed conversion ratio were the highest in SFO group than in FO group (p < 0.05). Fatty acid composition of fish body reflected the fatty acid composition determined in the experimental diets. The amounts of palmitic acid (PA; C16:0), oleic acid (OA; C18:1n-9), linoleic acid (LA; C18:2n-6) and docosahexaenoic acid (DHA; C22:6n-3) were dominant fatty acids in fish body. It was confirmed that the usage of safflower oil instead of fish oil in turbot feed did not generate any negative effects on growth, feed conversion and the values regarding the growth performance.  相似文献

Hematological and Immunological parameters of aquatic animals may be changes due to acute, subacute and chronic exposure to marine pollutants. The purpose of this study was to determine the experimental effects of water-soluble fraction (WSD) doses (0, 10, 100, 500 and 1,000?ppm) for 0, 48?h and 7?days on hematological and immunological features of juvenile great sturgeon Huso huso. Fish exposed after 48?h and 7?days showed a significant change in the white blood cell (P?<?0.01) in contrast to red blood cell (P?>?0.05). Only MCV, neutrophil and lymphocyte show significant change within 48-h exposure to WSD (P?<?0.05), whereas among significant indices, MCV and lymphocyte had been decreased and neutrophil had increased. Seven-day exposures showed a significant change in MCV, neutrophil, eosinophil and lymphocyte concentration in relation to the respective control (P?<?0.05), whereas among significant indices, neutrophils were significantly greater and MCV, eosinophil and lymphocyte were significantly lower than those in control groups (P?<?0.05).  相似文献

Sesamin (S) is a known lipid modulator and has been shown to increase the conversion of α-linolenic acid (ALA) to docosahexaenoic acid in rainbow trout (Oncorhynchus mykiss) fed vegetable oil mixtures including linseed oil. In this study, we evaluated the effects of S supplementation in linseed oil-based diets, content of α- and γ-tocopherols, fatty acid (FA) composition, as well as the gene expression of lipid-related genes. Fish with an average weight of 36.5 g were fed different combinations of commercial linseed oil (LO), purified linseed oil triacylglycerols (TAG) with polar fraction removed and a mixed linseed-sunflower oil (6:4 v/v) (MO). S was added at 0.58 g 100?1g feed and fed to the fish for a period of 58 days. Expression of PPARα was downregulated in white muscle of fish fed S containing diets (P < 0.05). The expression of PPARβ1A was not affected by S supplementation except where TAG oil was used. The expression of PPARβ1A declined significantly in TAG + S fed group (P < 0.05), which indicates that some minor compounds in linseed oil might suppress the effect of S on the expression of PPARβ1A. The expression of PPARγ(long) declined in LO + S and MO + S fed group (P < 0.05). The β-oxidation-related genes CPT1 and ACO were upregulated by vegetable oils compared to fish oil. S decreased percentage of ALA in white muscle of fish fed LO + S (P < 0.05). The increased desaturation index and the decreased ALA levels suggest that S may increase the biosynthesis of highly unsaturated FA in rainbow trout.  相似文献

This study investigated the effects of different soya oil products on growth, nutrient digestibility, and fatty acid composition of muscle in rainbow trout. The products’ ingredients were soap stock, fat powder, crude oil, and refined soya oil as a control. Four experimental diets were formulated by addition of 12 % of each of the four ingredients to a basal diet. Replacement of refined soya oil by the soya oil products did not influence growth performance until 8 weeks (P > 0.05). This replacement, however, reduced fat digestibility in rainbow trout (83.7 vs. 73.3–79.8 %; P < 0.05). Dietary inclusions of fat powder and crude oil led to larger values of protein digestibility than those of refined soya oil and soap stock (P < 0.05). The compositions of saturated and monounsaturated fatty acids of the muscle were similar, but linoleic acid concentration (LA, 18: 2n-6) increased in fish muscle fed refined soya oil compared to the other diets (37 vs. 30–31 %; P < 0.05). Arachidonic acid (ArA, 20: 4n-6) concentration was higher (P < 0.05) in fish fed both fat powder and soap stock than those fed the other diets. In conclusion, refined soya oil can be replaced by soya oil products in rainbow trout diet with no negative impacts on the growth. However, fish fed on diet containing refined soya oil had higher concentration of LA and lower density of ArA. It appears that rainbow trout is capable of digesting soya oil products though the digestibility rate reduced slightly with an increase in saturated fatty acid content.  相似文献

This study was performed to compare the various bioactivities of water-soluble, polar, and nonpolar extracts from the edible part (gonad) and waste (body wall) of the sea urchin Tripneustes gratilla. During antioxidant activity screening, these three extracts from both gonad and body wall showed various antioxidant activities in different tests. During human skin fibroblast (CCD966SK) viability and collagen screening, only water-soluble and polar extracts from gonad and body wall were observed to promote cell viability, while the water-soluble gonad extract promoted the collagen-generating activity of CCD966SK cells. The water-soluble and polar extracts from gonad and body wall showed proliferative activity towards different cell types. In contrast, the nonpolar extracts of gonad and body wall exerted antiproliferative effects on most tumor cells. These results indicate that the bioactivities of sea urchin extracts depend on the part of the urchin that the extract was taken from as well as the extraction method used.  相似文献

The objective of this study was to determine the effects of fish oil replacement with dietary vegetable oils on growth performance, chemical composition and fatty acids profiles in fillets of farmed Caspian great sturgeon juveniles Huso huso (26.97 ± 0.49 g). Five isonitrogenous and isolipidic diets were formulated, containing 10 % of added oil. The diet with 100 % kilka fish oil (Caspian tyulka, Clupeonella caspia) was the control. Fish oil was substituted by 50 % of vegetable oils consisting of an equal share of sunflower and soybean oils (diet A), sunflower and canola oils (diet B) and soybean and canola oils (diet C). In diet D, 100 % of fish oil was replaced with vegetable oil (1:1:1 ratio of sunflower oil, soybean oil and canola oil). Significant differences (P > 0.05) were not detected during 60 days feeding trial in final body weight, weight gain, condition factor, specific growth rate, feed conversion rate, protein efficiency ratio and the chemical composition of fillet (crude protein, crude lipid, moisture and ash). Generally, the fatty acids composition of fish fillets was reflective of the dietary lipid sources. These results indicate the feasibility of substituting fish oils with the mixture of vegetable oils in diets of juvenile H. huso without negative influence on growth providing optimum ratios of n-3/n-6 and n-3/18:1n-9 is met in the diet.  相似文献

The effects of acute temperature changes (2–17°C) on myocardial contractility with or without adrenergic activation were studied in the isolated spontaneously beating atrium of the Atlantic salmon (Salmo salar) reared at 8°C. The atrial frequency was markedly elevated (from 7 to 46 beats/min) by the rise in temperature from 2–17°C. Both the time to peak tension and to relaxation time were shortened. In contrast, the temperature effect on the maximal tension was modest. Exposure to exogenous adrenaline (1.1 nM–11 μM) resulted in a substantial enhancement of the maximal tension, notably at 2°C, while potentiation of the frequency at 2, 8 and 14°C, was less pronounced. The apparent affinity (pD2) for adrenaline on the chronotropy was higher at 8 and 14°C than at 2°C. For the inotropic responses pD2 was highest at the acclimation temperature (8°C). By comparison with data for the rainbow trout (Oncorhynchus mykiss) obtained by the same experimental design (Ask et al. 1981), species differences were apparent both in temperature dependence of contractile parameters and in their adrenergic activation. The Q10 for the frequency in absence of adrenaline was higher in the salmon than in the trout for the temperature interval 2–17°C. The apparent affinities for adrenaline for the frequency at 8°C and 14°C and for the maximal tension responses at 2°C and 8°C were also highest for the salmon atrium.  相似文献

A shortage of marine raw ingredients, such as fish oil, is predicted in the near future. The use of suitable alternative lipid sources, such as vegetable oils, is crucial for sustainable growth of the aquaculture sector. Three isonitrogenous (50% Crude Protein) and isoenergetic (19 kJ g−1) diets, in which fish oil was gradually replaced by soybean oil (0, 25 and 50% of the oil fraction) were tested for 12 weeks, using European sea bass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss) juveniles. No dietary effects (p>0.05) on growth performance, feed conversion, digestibility or body composition were observed with either species. The results obtained suggest the inclusion of soybean oil up to 50% of the dietary lipid is possible, in diets for sea bass and/or rainbow trout juveniles.  相似文献

This study was undertaken to assess the effects of fish oil (FO) substitution by a mixture of alternative vegetable oils (VO) on Seriola dumerili culture performance. A 154‐day feeding experiment was conducted using juveniles (39.2 ± 1.6 g average weight). Three isolipidic and isoenergetic meal‐based diets were formulated varying their lipid component. The control diet contained 100% FO (FO100), whereas diets VO50 and VO100 included 1/2 of oil blend and all the oil from blend of palm oil (PO) and linseed oil (LO) as substitute for FO, respectively. Dietary regime did not significantly affect growth performance, biometric indices, feed efficiency, plasma chemistry and liver and muscle lipid contents. Nonetheless, dietary VO inclusion impacted on the fatty acid profile of target tissues, especially in the liver. Fatty acid profiles of the fillets reflected those of the dietary oils except that there was apparent selective utilization of palmitic acid (C16:0) and oleic acid (C18:1n‐9) and apparent selective retention of long‐chain polyunsaturated fatty acids, especially eicosapentaenoic acid (EPA, C20:5n‐3) and docosahexaenoic acid (DHA, C22:6n‐3). The nutritional value and the potential ability to prevent the development of coronary heart diseases of the flesh lipid fraction decreased with gradual FO substitution.  相似文献

This experiment was carried out to determine the proximate composition of the mid-gut gland (MGG) of the Japanese scallop Patinopecten yessoensis to evaluate the characteristics of divalent minerals and water-soluble protein at various pHs, and to examine Cd-binding protein at different molecular weights. MGG of scallop contained protein, fat and ash of 28.9 g/100 g dry matter, 44.6 g/100 g dry matter and 6.78 g/100 dry matter, respectively. MGG also contained the macrominerals sodium, potassium, magnesium and calcium, and the trace minerals iron, zinc, cadmium and copper. The solubility of divalent minerals and water-soluble protein was high in both acidic and alkaline conditions, except that magnesium was not affected by acidity. The solubility of copper and iron had a positive correlation with water-soluble protein at all pH values, whereas cadmium had a strong correlation at alkaline pH. Low-molecular-weight water-soluble protein (fraction III, 437.5 < MW < 1,355) bound cadmium strongly in acidic, neutral and alkaline conditions. However, in acidic conditions cadmium had the strongest binding to protein.  相似文献

采用单细胞凝胶电泳技术(SCGE),以砂滤过的清洁海水为对照组,研究0~#柴油(0.015 mg·L~(-1)、0.03 mg·L~(-1)、0.06mg·L~(-1))和东海平湖原油(0.03 mg·L~(-1)、0.06 mg·L~(-1)、0.12 mg·L~(-1))在不同暴露时间(3 d、7 d、15 d和21 d)对黑鲷血细胞核DNA损伤作用。结果显示,黑鲷血细胞DNA损伤程度与两种石油有显著的剂量-效应和时间-效应关系。实验第3天,0~#柴油和东海平湖原油试验组就检测到DNA损伤,主要以1级损伤为主。随0~#柴油和平湖原油浓度的增高和胁迫时间的延长,彗尾DNA相对含量(Tail DNA%)和彗尾长度/核直径(TL/D)值均呈上升趋势,DNA损伤级别逐渐升高。恢复实验结束后,中、高浓度组0~#柴油(0.03 mg·L~(-1)、0.06 mg·L~(-1))和平湖原油(0.06 mg·L~(-1)、0.12 mg·L~(-1))的彗尾DNA相对含量(Tail DNA%)和彗尾长度/核直径(TL/D)值无法恢复至对照组水平(P0.05)。从DNA损伤程度来推测,0~#柴油的生物毒性大于平湖原油。研究表明,鱼类血细胞DNA损伤可作为监测海洋石油污染的生物标志物。  相似文献
