A single epithelium-free mammary fat pad was surgically prepared in each of twenty-five one-month-old, Friesian heifers. At 18 mo of age, heifers were randomly assigned to one of four treatment groups. Treatments were: control (C), growth hormone (GH), estrogen (E) or growth hormone + estrogen (GE). Hormones were administered for 40 hr before the animals were sacrificed to provide mammary samples of parenchyma (PAR), intact fat pad (MFP), and epithelium-free or "cleared" fat pad (CFP). IGF-1 and IGF binding protein-3 (IGFBP-3) mRNA was highest in CFP and MFP whereas the protein products were highest in PAR. IGFBP-2, a 28-kDa IGFBP and a 24-kDa IGFBP were more abundant in CFP and MFP. E and GH increased incorporation of [(3)H]thymidine into DNA of PAR. Incorporation of [(3)H]thymidine into the DNA of MFP or CFP was minimal. Coincident with the changes observed in mammary epithelial proliferation, E increased IGF-1 protein in MFP and PAR, and to a lesser extent in CFP. E tended to increase IGF-1 mRNA levels in MFP, but not CFP implying that the regulation of IGF-1 expression is modulated by adjacent epithelium. GH and E reduced IGFBP-3 protein in PAR and increased the 24-kDa IGFBP in CFP and MFP. Increased proliferation of mammary parenchymal cells was associated with increased IGF-1 and reduced IGFBP-3 protein in mammary tissue. An increase in the ratio of mammary IGF-1: IGFBP-3 likely increases the proportion of the mammary IGF-1 available to stimulate proliferation. These data also indicate that stromal: epithelial interactions regulate the IGF-1 axis in mammary tissue.  相似文献

Objective of the work was to study whether Mannheimia haemolytica may be transmitted from the mouth of the lambs into the teat of the dam during sucking. We compared bacterial populations within the teat duct and milk of ewes immediately before and immediately after sucking by the lambs. Tonsils of lambs of the ewes were swabbed. M. haemolytica strain DAG21T recovered from a teat duct of a ewe was compared to strain DAG21R recovered from the tonsils of her lamb by using 16s rRNA sequencing. We used those two isolates and another one of known pathogenicity, for challenging ewes: (i) 2-mm deep into healthy teats, (ii) 2-mm deep into teats with chapping lesions or (iii) into the cistern of healthy mammary glands. Of samples collected before suckling, 20/792 were bacteriologically positive, and of those after, 50/792 were bacteriologically positive (P < 0.001); in 37 cases, a negative sample became positive. One M. haemolytica (DAG21T) was recovered after suckling from a teat duct of a ewe. The organism was isolated from 57/90 tonsillar swabs from lambs. Risk of infection of ewe’ teats was 0.004 throughout lactation, being greatest (0.021) during the 3rd week of lactation. The 16s rRNA sequences of strains DAG21T and DAG21R were identical over 1450 nucleotides. Phylogenetic analysis showed that the two isolates clustered together with isolates of M. haemolytica. Organism deposition into healthy teats caused subclinical mastitis; deposition into teats with lesions or directly into mammary gland caused clinical mastitis. When results of inoculation of the three strains were compared between them, statistical significance was always P > 0.9. Results provide clear evidence that suckling by lambs can lead to transmission of M. haemolytica into the teats of the ewes; the bacteria have the potential to cause mastitis if circumstances are favourable.  相似文献

AIM: To compare clinical and ultrasonographic findings and assess the value of ultrasonography for evaluating the structure and condition of the teat and teat canal of sheep. METHODS: The mammary glands of 30 lactating ewes were examined clinically and using ultrasonography. The distance from the teat tip to the ground was measured and diagnostic images of the body of the gland, gland cistern and teat cistern were obtained using an 8.5 MHz frequency ultrasonic transducer. A California mastitis test (CMT) and bacteriological culture were performed on milk samples collected from each half of each udder. RESULTS: Udder parenchyma and teats were palpably normal in 27/30 (90%) ewes. Milk clots or abnormalities were evident in 4/30 (13%) udders, lesions in the glandular parenchyma and/or teats were evident by palpation in 3/30 (10%), and pathogenic bacteria were cultured from 2/30 (7%). In both of the latter cases, mastitis was diagnosed based on clotted and discoloured appearance of milk and highly positive CMT reactions. Ultrasonographic examination in these udders revealed non-homogenous regions in the glandular tissue and alternating areas of hyperechogenic and hypoechogenic tissue. Overall, highly positive CMT reactions were evident in milk from 14/30 (47%) left halves and 9/30 (30%) right halves (p=0.02). No relationship between teat-to-ground distances and CMT results was evident (p>0.05). Teat canal lengths ranged from 5.7-10.3 mm (mean=8.6; SD=1.3 mm) and the widths from 1.8-3.1 mm (mean=2.3; SD=0.4 mm). Milk sampled from ewes that had long (> or = 9 mm) or wide (> or = 2 mm) teat canals was highly positive to the CMT in 75% and 83% of cases, respectively. CONCLUSIONS: Ultrasonographic measurements of the length and width of the teat canal appeared to correlate with CMT results. The use of ultrasonography in evaluating the health of the udder of small ruminants holds promise for the future.  相似文献

In order to study the effects of sheep teat disorders on the protection of the mammary gland, we used a Mannheimia haemolytica isolate, which did not cause clinical mastitis when deposited into intact teats. In the first experiment, this was deposited into the duct of teats with orf (Group A, n=5) or papilloma (Group B, n=3). In the second, teats were chapped and then, the organism was deposited into the duct (Group C, n=7) or on the skin (Group D, n=4). Ewes with healthy teats were controls (Group E, deposition into duct, n=5; Group F, deposition on skin, n=2). The ewes in Groups A, B or C developed clinical mastitis 5 h later, whilst the ewes in Group D developed it 2 d later; no control ewe developed clinical mastitis. In ewes with teat lesions, the organism was isolated from secretion samples and the California Mastitis Test became positive 5 h after challenge; neutrophils and lymphocytes were seen in Giemsa-stained secretion films from Group A or B ewes, whilst macrophages, neutrophils and lymphocytes in films from Group C or D ewes; neutrophils were predominating in films from Group E or F ewes. Inside the teats of Group A, B, C or D ewes, folds, hyperaemia and mucosal thickness were seen; histologically, subepithelial leucocytic infiltration was seen. In Group A or B ewes, no evidence of lymphoid tissue at the teat duct-cistern border was found. In Group C or D ewes, intense erosion and ulceration of the teat skin and conspicuous lymphoid tissue at the teat duct-cistern border, were evident; lesions characteristic of haemorrhagic mastitis were in the mammary parenchyma. In control ewes, subepithelial leucocytic infiltration in the teat duct and lymphoid tissue as above, were evident. We postulate that teat lesions can be predisposing factor to mastitis, by adversely affecting defences and speeding the process of infection and making it more severe.  相似文献

A knowledge of the microbiological status of milk and of the different structures in the mammary glands has great importance in elucidating the pathogenesis of mammary gland infections. The objective of this study was to evaluate the microbiological status of various structures in the mammary glands from naturally infected dairy cows following slaughter. A total of 94 samples of milk, 184 samples of mammary parenchyma, 168 samples of gland cisterns, and 168 samples of teat cisterns were collected for microbiological examination. Microorganisms were detected in 59.9% of all samples, 67.0% of the milk samples, 70.1% of the mammary parenchymas, 55.9% of the gland cisterns and 48.8% of the teat cistern samples. When all samples were considered, coagulase-negative Staphylococcus were the most prevalent (35.7%) followed by coagulase-positive Staphylococcus (12.2%), Corynebacterium bovis (2.4%), Prototheca sp. (1.9%), and Streptococcus dysgalactiae (1.5%). There was a significantly higher occurrence of microorganisms in the milk and mammary parenchyma compared to the gland cisterns and teat cisterns. We describe the features of 13 cases of teat disorders in ewes, as follows: lengthy teats (one case), papilloma infection (two), staphylococcal dermatitis (one), nodules and abscesses (two), teat bites (three), stenosis (one), orf (two), compressed teat (one). Mannheimia haemolytica, Staphylococcus epidermidis, Staphylococcus simulans or Staphylococcus aureus were isolated from samples. Sub-epithelial leucocytic infiltration at the teat duct-teat cistern border, but not lymphoid nodules, was the salient histological finding in the teat. Leucocytic infiltration and destruction of parenchyma were evident in the mammary gland. The results provide field evidence suggesting that teat lesions predispose ewes to mastitis.  相似文献

Effects of number of lambs suckled and supplemental protein source on ewe performance were determined. Lactation performance of ewes nursing twin or triplet lambs was compared in Exp. 1 and 2. In Exp. 1, ewes giving birth to and nursing triplets produced 21% (P less than .05) more milk/unit metabolic body weight, were 10% more efficient in the conversion of feed to milk and produced 26% (P less than .05) more kg of lamb/ewe on d 42 of lactation than did ewes giving birth to and nursing twins. In Exp. 2, lactation performance of ewes giving birth to triplets and rearing only two lambs was compared with performance of ewes giving birth to triplets and rearing three lambs. Ewes rearing three lambs produced 28% (P less than .08) more milk and were 26% (P less than .05) more efficient in converting feed to milk than were those rearing two lambs. This suggests that direct nursing stimulation of the mammary gland is responsible for the improvement in milk production observed when ewes nurse additional lambs. The efficacy of slowly degradable protein sources for improved lactation was studied in Exp. 3 and 4. When blood meal was fed to twin-rearing ewes at 3.30% of the diet (25% of total dietary protein), milk production was greater (3,176 vs. 2,506 g/d, P less than .05) and efficiency of milk production was greater (1.05 vs .79 g milk/g feed, P less than .05) than when ewes were fed supplemental soybean meal. Subclinical mastitis was induced by inoculating one mammary gland in dairy ewes (n=8) with a Staphylococcus simulans isolate; control ewes (n=4) were included. The milk yield of inoculated glands decreased (P<0.001), the California Mastitis Test (CMT) score increased and the organism could be recovered from the inoculated glands. With time, there was significantly increased frequency of "hindering sucking" (P=0.016) and "head up posture" (P<0.001) in the control ewes. Infected ewes had a significantly increased frequency of "vocalisation" (P=0.013) compared to controls. There was a significant difference in the frequency of "sucking attempt" and "successful suck" (P<0.05) behaviours between lambs of the two groups. Lambs of the challenged ewes also showed significantly increased frequency and duration of these behaviours towards the uninoculated glands of their dams, rather than to the challenged glands (P<0.05); no such difference was evident for the lambs of control ewes. It was concluded that subclinical mastitis alters the sucking behaviour of both ewes and lambs.  相似文献

We aimed (i) to determine differences in bacterial flora of teat duct and mammary gland of ewes before and after suckling, (ii) to evaluate factors potentially affecting those. We collected samples of teat duct material and mammary secretion from 11 ewes immediately before and after sucking by lambs, as well as 120 min later. We processed samples bacteriologically and compared changes in infection by the Sign Test. We isolated bacteria from 3.5% duct and 1.5% secretion samples before suckling. Respective figures post-suckling were 10.6% and 2.0%, and 120 min later 6.8% and 1.5%. We recorded differences in infection of duct samples before and after suckling in 40 cases; bacteria were isolated before suckling from six samples, whereas after it from 34 (p < 0.001). Also, we recorded differences in samples collected after suckling and 120 min later in 12 cases; bacteria were isolated immediately post-suckling from eight samples, whereas 120 min later from four (p = 0.375). No significant changes were seen for secretion. We found neither difference between ewes with single or twin lambs, nor among stages of lactation. Mostly, we isolated staphylococci: 70% of isolates before suckling, 80% of isolates after it, 91% of isolates 120 min later. After suckling we also isolated two Mannheimia haemolytica strains. Suckling predisposes to entrance of bacteria into the teat; however, increased teat infections did not result in mammary infections. Isolation of M. haemolytica post-suckling indicates that lambs act as source of infection for this pathogen.  相似文献

Seven out of eight ovine ureaplasma strains inoculated into the mammary gland of suckling ewes produced a mastitis. The pattern of infection was single phase in 5 ewes, persisting for 12-41 days, and biphasic in 3 ewes, persisting in 2 of them until weaning at 60 days and 3 months post-infection. Sucking lambs did not become infected in the eye or nasal areas, and did not transfer infection to the control contralateral glands.  相似文献

Data were collected from intact males, castrated males and ewe lambs to investigate the effect of presence or absence of testosterone prenatally and during the postweaning period on postweaning growth, feed intake and carcass chemical composition. Half the lambs from each sex were the progeny of dams that had received five injections of testosterone cyprionate from d 32 through d 87 of gestation. Linear contrasts were used to detect differences. Postweaning daily gain of intact males was greater (P less than .01) than that of male castrates. Ewe lambs from treated dams had approximately 12% greater rate of growth (P less than .04) than ewe lambs from control dams. Ewe lambs from dams that had been treated were 28% more efficient (P less than .01) in the conversion of food to weight than those from untreated dams. Ewe lambs from treated dams had heavier livers (P less than .07). Carcass protein for intact males was greater (P less than .11) than for castrates, and extractable fat was less (P less than .05). Masculinization of growth characteristics of ewe lambs affected the quantity of carcass fat relative to control ewes (7.59 vs 8.92 kg). These ewe lambs also had more water in the carcass than did the control ewes (13.93 vs 12.29 kg). Administration of exogenous testosterone to pregnant ewes over an interval of time approximating time of sexual differentiation in the fetus enhances postweaning growth rate, feed conversion efficiency and chemical composition of genetic females.  相似文献

To examine effects of nutritional plane and Se supplementation on colostrum quality and mammary development, individually fed, pregnant Rambouillet ewe lambs were allotted randomly to 1 of 6 treatments in a 2 x 3 factorial arrangement. Main effects included dietary Se level, which began at breeding (d = 0) [adequate Se (9.5 mug/kg of BW) vs. high Se (81.8 mug/kg of BW)], and plane of nutrition, which began at d 50 of gestation [60% (RES), 100% (CON), and 140% (HIGH) of requirements]. Upon parturition, lambs were immediately separated from dams and weighed. Three hours after lambing, colostrum yield was determined, and samples were obtained for components and immunoglobulin G (IgG) analysis. Ewes were slaughtered within 24 h of parturition, and mammary tissues were collected for determination of alveolar secretory epithelial cell proliferation index and luminal area. Gestation length was reduced (P < 0.01) in HIGH ewes compared with RES and CON ewes. Although birth weights were reduced (P < 0.01) in RES and HIGH compared with CON ewes, there was little effect of diet on placental size. Mammary gland weight was reduced (P /= 0.15) on mammary gland weight, colostrum quantity, or IgG concentration in pregnant ewe lambs. Improper nutrition from mid to late pregnancy in ewe lambs altered colostrum quality and quantity and reduced offspring birth weight, which may have negative implications for lamb health and survival during the early postnatal period.  相似文献

Effects of estrogen and progesterone on the first abdomino-inguinal mammary gland and the associated blood vessels in ovariectomized mice were investigated morphometrically, and light and electron microscopically. Although there were no significant differences in the area of the fat pad of the mammary gland among the experimental groups, the area of the mammary parenchyma and diameters of the blood vessels supplying the mammary gland, i.e., A. et V. circumflexa ilium profunda and A. et V. epigastrica caudalis superficialis, reached the maximal value in ovariectomized mice treated with estradiol (E) + progesterone (P). Similarly, the blood capillaries around buds and ducts of the mammary gland were most densely distributed in E + P treated mice. In the adipose tissues of the mammary stroma in intact mice, fat cells were of multilocular type in the peripheral regions around the main vessel, and of unilocular type in the other part. In E and E + P treated mice, however, fat cells were mostly of unilocular type. These findings suggest that the formation of mammary fat tissues may occur in advance of that of the mammary parenchyma. By TEM, epithelial cells of the mammary parenchyma contained a large number of mitochondria and ribosomes, well-developed Golgi apparatus and rER, and large lipid droplets. Endothelial cells of blood capillaries displayed numerous pinocytotic vesicles, longer marginal folds and microvillous processes. Ewes treated by intramammary infusion of cephapirin benzathine at the end of lactation were less likely to develop new intramammary infections by early in the next lactation. A controlled clinical trial involving 135 treated ewes and 145 untreated control ewes evaluated the prophylactic and treatment efficacy of intramammary antibiotic treatment of ewes at the time of weaning lambs. Milk samples for bacteriologic examination and identification of intramammary infections were collected at weaning of lambs and 1 to 3 weeks into the subsequent lactation. Untreated ewes were 2.6 times more likely than treated ewes to develop new intramammary infections between sampling times. Existence of an intramammary infection in one mammary gland significantly increased the risk of the other mammary gland becoming infected. Treatment also appeared to be associated with a higher cure rate of existing infections.  相似文献

We studied the possible effects of bacterial populations within the teat duct, in the pathogenesis of ovine mastitis. In experiment I, 32 ewes were allocated into group A (ewes from which we isolated (+++ growth) coagulase-negative staphylococci), B (ewes from whose duct we isolated (+ growth) coagulase-negative staphylococci) or C (ewes from which we isolated Bacillus spp.) and subdivided into A1, B1, C1 (n=4; challenged by deposition of 1.250 cfu of Mannheimia haemolytica into the teat duct) or A2, B2, C2 (n=4; used as uninoculated controls); group D (n=8) contained ewes with no bacteria in their teat ducts and were challenged as above. There were less bacteriological isolations of flora (P = 0.018) and challenge (P<0.05) organisms from A1 than from A2 and D ewes; the severity of pathological findings in A1 (summed up score: 27) ewes was smaller than in D (summed up score: 36) ewes (P = 0.038). No such findings were evident with B1 or C1 ewes (P>0.4). In experiment II, ewes (groups E and F, n=6) from whose duct we isolated coagulase-negative staphylococci (+ growth) were used; in group G (n=6) ewes with no bacteria in their teat ducts were included. Teat chapping was applied in E and G ewes. All E ewes developed acute clinical mastitis within 24 h after teat chapping, although we had carried out no challenge; there were more bacteriological isolations of flora organisms from E than from F and G ewes (P < 0.001); the severity of pathological findings in E (score: 28) was greater than in F (score: 3) or G (score: 14) ewes. In experiment III, eight ewes with no bacteria in their teat ducts were allocated into group H or I (n=4) and challenged into the teat (group H) or into the gland (group I) with 10(6) cfu of a Staphylococcus simulans recovered from the teat duct of a group E ewe. Group H ewes developed transiently clinical followed by subclinical mastitis (based on bacteriological and cytological evidence), whilst group I ewes developed severe clinical disease. We conclude that staphylococcal flora present in high numbers within the teat duct of ewes can afford some protection against invading microorganisms. However with impeded defence mechanisms of the teat, the same flora may invade the mammary parenchyma and cause clinical mastitis.  相似文献

Mature pregnant crossbred ewes (n = 90) were used in a randomized complete block design and assigned to 1 of 3 winter-feeding systems differing in primary feed source: haylage (HL), limit-fed corn (CN), or limit-fed dried distillers grains (DDGS). Effects of these winter-feeding strategies on ewe and lamb performance were determined. Diets were formulated to meet or exceed NRC (1985) nutrient requirements during gestation and were fed from about d 60 of gestation until parturition. All ewes were fed a common diet postpartum. Every 2 wk during gestation, BW and BCS were collected and diets were adjusted to maintain similar BW gain for ewes fed CN and DDGS vs. HL. At 80 and 122 d of gestation, jugular blood samples were collected at 0, 3, 6, and 9 h postfeeding to measure plasma glucose, insulin, NEFA, and blood urea nitrogen concentrations. At birth, 6 lambs per treatment were killed to measure body composition. At 28 ± 2 d postpartum, milk yield was measured. Lambs were weaned at 61 ± 4 d of age. During mid gestation (d 60 to 115), BW gain of ewes was similar among treatments; however, at d 115 of gestation ewes fed HL had a smaller (P = 0.04) BCS than ewes fed DDGS or CN. Plasma glucose concentrations were greater (P ≤ 0.004) in ewes fed CN than in those fed HL or DDGS just before feeding on d 80 and 122 of gestation, whereas ewes fed DDGS vs. CN or HL had greater (P ≤ 0.04) plasma insulin concentrations at 3 h postfeeding. At parturition, ewe BW was greatest for DDGS, least for HL, and intermediate for CN (P ≤ 0.003). Ewes fed CN and DDGS had greater BCS at parturition than those fed HL, but by weaning, ewes fed DDGS had greater BCS (P ≤ 0.05) than those fed CN or HL. Birth BW tended (P = 0.09) to be heavier for lambs from ewes fed CN and DDGS than from those fed HL prepartum, but there was no difference (P = 0.19) due to ewe gestation diet on lamb BW at weaning. At birth, lamb muscle, bone, organ, and fat measures were not affected (P > 0.13) by treatment. Ewe milk production and lamb preweaning ADG were also similar (P > 0.44) among treatments. Prepartum dam winter feed source did not have detrimental effects on pre- or postpartum ewe performance, but altered prepartum maternal nutrient supply during gestation, which affected birth weight but not preweaning growth or mortality.  相似文献

Pregnant ewes (large frame [LF] and small frame [SF]) were nutritionally stressed in early gestation (EGS), late gestation (LGS) or fed 100% of NRC requirements (unstressed, US) throughout gestation. Lambs (128) from these ewes were slaughtered at birth, weaning (18 kg), 41 kg or 55 kg. Sixty-four lambs received a 13% protein diet from weaning to either 41 or 55 kg. Lambs from SF ewes were fatter at 55 kg, had a higher numerical yield grade and a lower percentage of carcass protein. Lambs from US ewes were youngest at slaughter and had the most carcass weight and protein per day of age at 55 kg. The LGS lambs had the lowest percentage of lean and carcass protein at 41 and 55 kg. However, at birth these lambs had the highest concentration of RNA and DNA in muscle. The EGS lambs had the lowest quality grade, carcass weight per day of age and fat percentage. Muscle DNA and RNA at birth was lowest in EGS lambs. However, EGS lambs produced the highest lean percentage and highest percentage carcass protein at 41 and 55 kg. Shortest metacarpals and metatarsals were also found in these lambs at weaning and 41 kg. Although frame size had little effect on carcass characteristics, the effects of nutritional stress in the first 80 d of gestation were apparent in lambs slaughtered at 31 kg. Stress in the last 50 d of gestation had more effect on lambs slaughtered at 55 kg.  相似文献

The influence of plane of nutrition, growth hormone (GH) treatment and dietary polyunsaturated fat on serum concentrations of GH and insulin (INS) and binding capacities of GH, INS and prolactin (PRL) in liver, mammary parenchyma and adipose tissue was assessed in prepubertal ewe lambs. Ten lambs were assigned to each of four treatment groups. Treatments included: (A) lambs with ad libitum access to a high-energy ration; (G) lambs fed as group A and treated with bovine GH (.08 mg/kg/d); (R) lambs with feed intake restricted to limit ADG to about 120 g; and (S) lambs with ad libitum access to a ration including formaldehyde-protected sunflower seed. Diets, all approximately isonitrogenous and isocaloric, were fed from about 7 to 22 wk of age. Weekly blood samples were collected during wk 6 to 14 of the trial. Averaged across sampling dates, mean serum concentrations of GH were elevated in G lambs (P less than .05) and INS concentrations differed in the order G greater than A greater than R = S (P less than .05). Crude membranes for binding assays were prepared from liver, mammary parenchyma and adipose tissue. Mean concentrations of GH receptors in liver and PRL receptors in mammary parenchyma were elevated in group S lambs (P less than .01). Dietary polyunsaturated fat increased the number of GH receptors in liver and PRL receptors in mammary parenchyma. Increased availability of receptors may mediate the stimulation of mammary growth observed in lambs fed polyunsaturated fat.  相似文献
