AIM:To compare the effects of carvedilol, cilazapril and their combination on left ventricular remodeling(LVRM) after acute myocardial infarction(AMI) in rats. METHODS: Twenty-four hours after AMI operation, 100 surviving rats were randomly assigned to: ①AMI control(n= 25), ②AMI+carvedilol(1 mg·kg-1 ·d-1, n= 25)(C1), ③AMI+cilazapril(1 mg·kg-1 ·d-1, n= 25)(Z1), and ④ AMI+combination(n= 25) groups. Sham-operated group(n= 17) were selected randomly. After 4 weeks of therapy with the drugs gastric gavage, hemodynamic and pathological studies were performed. RESULTS: There were no significant differences in MI size among the four AMI groups(all P> 0.05) Left ventricular(LV) end diastolic pressure(LVEDP), volume(LVV), weight(LVW) and septal thickness(STh) were all higher and left ventricular pressure maximal rate of rise and fall(±d p /d t) were lower(all P< 0.01) in AMI group than sham-operated group. The LVEDP, LVV, LVW and STh were all lower and ±dp /dt were higher in Z1, C1, and combination groups than those in AMI group(P< 0.05, P< 0.01), with LVEDP and STh were more lower in the combination group than in the two monotherapy group(P< 0.05, P< 0.01), but there were no significant differences in other variables among the three therapy groups. CONCLUSION: Carvedilol, cilazapril and their combination all can prevent from LVRM after AMI in rats, improve hemodynamics and LV function, with the combination superior.  相似文献

AIM: To investigate the cardiac AMP-activated protein kinase (AMPK) activity and the effects of AMPK activator on cardiac structure and function in the mice with different β-adrenoceptor (β-AR) stimulation patterns. METHODS: Male BALB/c mice were subcutaneously injected with AMPK activator (AICAR, 250 mg· kg-1·d-1) or saline, and infused with β-AR agonist isoproterenol (ISO, 5 mg·kg-1·d-1) for 14 d. The cardiac functions were evaluated by echocardiography or hemodynamic method, and the hearts were harvested after infusion cessation immediately or 3 d later. Phosphorylated AMPK (p-AMPK) was measured by Western blot. RESULTS: Sustained ISO infusion increased p-AMPK level. AICAR did not further increase p-AMPK but attenuated ISO-induced increase in heart weight. Sustained ISO infusion increased cardiac systolic function as indicated by left ventricular fractional shortening (FS) and maximum rate of pressure rise (+dp/dtmax). The cardiac systolic function was not further increased by AICAR. The cardiac diastolic function as indicated by left ventricular end-diastolic pressure (LVEDP) was not different in each group. In contrast, cardiac p-AMPK level was similar between the control mice and the mice with sustained ISO infusion and ceased infusion for 3 d. In this model, AICAR improved the cardiac systolic and diastolic functions, which were impaired by ISO. Moreover, the increased pattern of p-AMPK level was similar with that of heart rate upon ISO stimulation. CONCLUSION: Sustained ISO infusion increases p-AMPK. After ISO infusion cessation for 3 d, p-AMPK is decreased to the basal level. β-AR-induced inotropic effects should be avoided to investigate the cardioprotective role of AMPK activation in the β-AR stimulation models.  相似文献

AIM: To investigate the effects of long-term TCV116 on left ventricular remodeling and heart function after myocardial infarction. METHODS: Myocardial infarction (MI) was caused by ligation of the left anterior descending coronary artery in rats. One week after the surgical performance, the surviving rats were randomly assigned to the following treatment protocols: (1) MI rats with no therapy; (2) MI rats treated with TCV116 2 mg/kg per day; (3) Sham-operated control; (4) Sham-operated rats, treated with TCV116 2 mg/kg per day. At 22 weeks, cardiac hemodynamic parameters such as MAP, LVSP, dp/dtmax and LVEDP, and histomorphometric parameters such as LVW/BW and LVCA/BW were measured, mRNA of cardiac genes such as βMHC, BNP, TGF-β1, collagen I and III were quantified, and survival rates were calculated. RESULTS: Compared with sham-operated rats, MI rats without therapy showed significant increases in histomorphometric parameters as well as in mRAN expressions of cardiac genes (P<0.01); While their hemodynamic parameters were significantly impaired (P<0.01), and survival duration shortened (P<0.05). Compared with MI rats without therapy, MI rats treated with TCV116 showed significant attenuation of mRAN expression of cardiac genes (P<0.01); While their hemodynamic parameters were significantly improved (P<0.05 or P<0.01), and survival duration extended (P<0.05). CONCLUSION: Treatment with long-term angiotensin II type 1 receptor antagonist may improve left ventricular remodeling and cardiac function after MI in rats.  相似文献

AIM: To investigate effect of leonurine on the expression of microRNA-1 (miR-1) in rats with myocardial fibrosis induced by isoproterenol (ISO). METHODS: SD rats (n=10) were used as normal control group, and 80 rats were given ISO by intraperitoneal injection daily for 2 weeks to establish the model of myocardial fibrosis. The model rats were randomly divided into 5 groups:model group, low-dose (7.5 mg·kg-1·d-1) leonurine group, middle-dose (15 mg·kg-1·d-1) leonurine group, high-dose (30 mg·kg-1·d-1) leonurine group and p38 mitogen-activated protein kinase (p38 MAPK) inhibitor (0.3 mg·kg-1·d-1) group. After the treatment for 2 weeks, the ultrastructure of left ventricular myocardial tissues was observed under electron microscope. Masson staining was used to detect collagen fibrosis, and the expression of collagen I and collagen Ⅲ was determined by the method of immunohistochemistry. The contents of endothelin-1 (ET-1) and angiotensin Ⅱ (Ang Ⅱ) were measured by ELISA. The expression of miR-1 and ET-1 mRNA was detected by real-time PCR, and the protein expression of p38 MAPK, β-myosin heavy chain (MHC) and α-MHC was determined by Western blot. RESULTS: Compared with model group, the ultrastructure of left ventricular myocardial tissues in high-dose leonurine group was attenuated, and the expression of miR-1 and the protein expression of α-MHC in left ventricular myocardial tissues of high-dose leonurine group were increased (P<0.05). Collagen volume fraction, collagen I, collagen Ⅲ, the ratio of collagen Ⅰ/collagen Ⅲ, the contents of ET-1 and Ang Ⅱ, the mRNA expression of ET-1, and the protein expression of p38 MAPK and β-MHC in high-dose leonurine group were lower than those in model group (P<0.05). CONCLUSION: Leonurine attenuates myocardial fibrosis in the rats induced by ISO, and it is potentially associated with affecting the expression of miR-1, and inhibiting ET-1/p38 MAPK signaling pathway.  相似文献

AIM: To study the effects of metformin on the pressure overload-induced cardiac hypertrophy in rats. METHODS: Transverse aortic constriction (TAC) model of rat was made through laparotomy. One week after TAC surgery, the rats were randomly divided into 5 groups (n=8 in each group) and were administered with the corresponding drugs orally every day for 8 weeks: sham group (sham surgery, administered with 2 mL distilled water); TAC group (TAC rats, administered with 2 mL distilled water); metformin (MET) group (TAC rats, administered with MET at dose of 300 mg·kg-1·d-1); MN group [TAC rats, administered with MET at dose of 300 mg·kg-1·d-1 plus NOS inhibitor, NG-nitro-L-arginine methyl ester (L-NAME) 50 mg·kg-1·d-1] and L-NAME group (TAC rats, administered with L-NAME at dose of 50 mg·kg-1·d-1). After treated for 8 weeks, the echocardiography, hemodynamics, the ratio of heart weight to body weight (HW/BW) and histological examination of the heart were performed. The levels of myocardial AMP-activated protein kinase subunit α (AMPKα), p-AMPKα Thr172, endothelial nitric oxide synthase (eNOS) and p-eNOS Ser1177 were detected by Western blotting. Plasma and myocardial nitric oxide (NO) were detected biochemically. RESULTS: After 8 weeks treatment, the wall thickness of left ventricle, the heart weight/body weight ratio (HW/BW), and the left ventricular myocardial perivascular fibrosis and myocardial interstitial fibrosis of the animals in TAC group were significantly increased as compared to those in sham rats. Treatment with MET for 8 weeks significantly attenuated left ventricular hypertrophy and improved cardiac function in TAC rats. These effects of MET were mostly abolished by L-NAME. Molecular biology and biochemical testing revealed that the levels of left ventricular myocardial p-AMPKα Thr172 and p-eNOS Ser1177, as well as the levels of myocardial and serum NO were significantly increased in MET group. CONCLUSION: Long-term MET treatment significantly inhibits the cardiac hypertrophy and the myocardial fibrosis and improves the cardiac functions in pressure-overload rats. The anti-hypertrophic effects of MET may be mediated via activation of AMPK-eNOS signaling pathway.  相似文献

AIM: To observe the effects of pioglitazone on myocardial energy metabolism and hemodynamics in rats with heart failure after myocardial infarction (MI). METHODS: The model of MI was established by ligation of left anterior descending artery. The 20 surviving rats were randomly divided into MI group (n=10) and pioglitazone intervention group (P group,n=10, pioglitazone 3 mg·kg-1·d-1 orally). The sham-operated rats (SH, n=10) served as controls. Hemodynamic parameters were measured. The ratio of left ventricular weight to body weight (LVW/BW) and the ratio of right ventricular weight to body weight (RVW/BW) were calculated after 8-week treatment. The expression of PPARγ was examined by Western blotting. Mitochondrial respiratory function was determined by Clark oxygen electrodes. The size of adenine acid pool (ATP, ADP and AMP) in mitochondria was measured by HPLC. The adenine nucleotide translocator(ANT) activity was detected by the atractyloside-inhibitor stop technique. RESULTS: Compared with SH group, the protein expression of PPARγ was significantly decreased in MI group (P<0.01). The mitochondrial respiratory activity, the transport activity of ANT and the high-energy phosphate content were decreased in MI group (P<0.01), and the hemodynamic parameters were in disorder (P<0.01). Compared with MI group, the protein expression of PPARγ in P group was significantly increased. The mitochondrial respiratory activity, the high-energy phosphate content, the transport activity of ANT were improved (P<0.01). However, the hemodynamic parameters were not significantly changed.CONCLUSION: Pioglitazone increases the protein expression of PPARγ and improves myocardial energy metabolism in the development of heart failure in the rat model of myocardial infarction, but dose not change the hemodynamic parameters significantly.  相似文献

AIM: To investigate the effect of rhynchophylline (Rhy) on blood pressure, cardiac hypertrophy and myocardial fibrosis in spontaneously hypertensive rats (SHR). METHODS: Spontaneously hypertensive rats were randomly divided into model group, high dose (10 mg·kg-1·d-1) and low dose (2.5 mg·kg-1·d-1) group of rhynchophylline, captopril group (17.5 mg·kg-1·d-1). Wistar-Kyoto rats were used as normal control. Respectively, systolic blood pressure was measured by tail cuff every 2 weeks. After 10 weeks, heart weight index and left ventricular weight index were calculated. The myocardial hydroxyproline and plasma angiotensin Ⅱ were detected. Moreover, basic myocardial histopathological changes and myocardial collagen fibres were observed by HE staining and Masson staining, respectively. The protein expression of TGF-β1 and Smad3 in the myocardium was measured by the methods of immunohistochemistry and Western blot. RESULTS: Compared with SHR model group, Rhy significantly reduced blood pressure (P<0.05), the levels of HYP in the myocardium (P<0.05) and the levels of AngⅡ in the plasma (P<0.01). The pathological damages of the myocardial tissues and collagen deposition were attenuated. The protein expression of TGF-β1 and Smad3 was significantly reduced by the treatment with Rhy (P<0.01). CONCLUSION: Rhynchophylline reduces blood pressure and adjusts to improve ventricular remodeling of SHR. The mechanism may be involved in the TGF-β1/Smad pathway and reducing AngⅡ content.  相似文献

AIM: To investigate the effects of Qinghao-Biejia (QHBJ) decoction on ovarian cancer in female rats by dimethylphenyl allium. METHODS: Ovarian cancer female rat model was established by embedding medicine thread with dimethylphenyl allium in situ. After successful modeling, the rats were divided into model group with the same volume of normal saline, high dose group with QHBJ decoction at 3 g·kg-1·d-1, medium dose group with QHBJ decoction at 1.5 g·kg-1·d-1, low dose group with QHBJ decoction at 0.75 g·kg-1·d-1 and western medicine group with meloxicam at 4 mg·kg-1·d-1. After 6 weeks of administration, the tumor weight and tumor inhibition rate of ovartian tumor were measured.Expression of Bcl-2 and Bax proteins were detected by immunohistochemistry. The protein levels of AKT, p-AKT and BTAK were determined by Western blot. RESULTS: QHBJ decoction decreased the protein levels of Bcl-2, AKT, p-AKT and BTAK, increased the protein level of Bax,and reduced the tumor weight of ovarian tumor. The anti-tumor rate of ovarian tumors in each group showed a significant increase in a dose-dependent manner with QHBJ decoction (F=34.2, P=0.000 5). CONCLUSION: QHBJ decoction inhibits the proliferation of ovarian cancer cells induced by dimethylphenyl allium by regulating the balance of Bcl-2 protein and Bax protein, decreasing the protein levels of AKT, p-AKT and BTAK.  相似文献

AIM: To investigate the influences of electric stimulation applied during the absolute refractory period (ARP) on the cardiac function of normal rabbits and rabbits after myocardial infarction (MI) and to observe the regional effects of this electric stimulation. METHODS: 64 rabbits were randomly assigned to normal and MI groups and each group was then divided into the anterior and posterior groups. A thoracotomy was performed 4 weeks after MI in rabbits. One set of electrodes was inserted into the anterior and posterior wall of left ventricle of the anterior and posterior groups, respectively. Current pulses were delivered during the ARP (called CCM) during sinus rhythm in rabbits. The left ventricular systolic pressure (LVSP) and the left ventricular end diastolic pressure(LVEDP) as well as maximum positive and negative left ventricular pressure change (±dp/dtmax) were observed. RESULTS: In the normal and MI groups, LVSP, +dp/dtmax significantly increased, and LVEDP, -dp/dtmax were reduced during CCM stimulation compared with the baseline (P<0.05). In the normal rabbits, electric stimulation in the anterior group improved the cardiac function more significantly than that in the posterior group (P<0.05). In the MI rabbits, there was no difference between the anterior and the posterior groups (P>0.05). CONCLUSION: Electric stimulation delivered during the ARP significantly enhances the contractility and the relaxation of myocardium in normal rabbits and rabbits after MI, and the effects of CCM stimulation on heart are regional.  相似文献

AIM: To clarify the protective effect of long-term administration of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitor fluvastatin on ventricular remodeling after myocardial infarction (MI) in rats and its mechanisms. METHODS: Myocardial infarction were established by ligated left coronary anterior artery in SD rats, 24 hours after the operation, the survival rats were treated by gavage fluvastatin (20 mg·kg-1·d-1) or distilled water for 8 weeks. Doppler echocardiography, homodynamic and cardiac histomorphometry were used to assess the ventricular remodeling and cardiac function. The plasma levels of total cholesterol (Tch), creatinine (Cr), glutamic-oxal (o) acetic transaminase (AST), lipid peroxidation (LPO), glutathione perioxidase (GSH-PX), nitrogen monoxide (NO2-/NO3-) were detected. RESULTS: The Tch, Cr and AST were not significant difference in groups. Left ventricular end-diastole pressure, right relative weight, left ventricular posterior wall thickness, collagen volume fraction and the lung weight were decreased in AMI+fluvastatin group compared to AMI group (P<0.05, P<0.01); The levels of LPO, NO2-+NO3- in plasma and LPO in myocardium decreased, but plasma GSH-PX level increased in AMI+fluvastatin group (P<0.05). CONCLUSION: Fluvastatin ameliorates the ventricular structural remodeling in a rat model of infarction, and delays the development of heart failure. The anti-oxidation mechanism of fluvastatin may take part in this process.  相似文献

AIM:To reveal the pathogenesis of salt-induced hypertension.METHODS:Forty male SD rats were divided into five groups, which received on same chow but different drink. Control(NC)group: deionized water; High salt(HS)group: 1.5% NaCl solution; L-arginine(HS+Arg)group: L-arginine(4 g·kg-1·d-1)in 1.5% NaCl solution; Enalapril (HS+En) group: enalapril (30 mg·kg-1·d-1) in 1.5% NaCl solution; Terazozin(HS+Ter)group: terazozin(4 mg·kg-1·d-1)in 1.5% NaCl solution. At the end of 8 weeks, rats were anesthetized with pentobarbital sodium. Blood pressure(BP)were recorded and blood were drawn from inferior vena cava and kidneys, adrenals were removed. NO(x),ET and AngII, Na-K-ATPase and proscillaridin-like compound(PLC)were assayed.RESULTS:BP, PLC and ET in plasma and AngII in adrenal were increased, NO(x)and AngII in plasma and kidney were decreased in HS group compared with NC group.CONCLUSION:High salt intake may induce hypertension in SD rats. In addition to the Na-K-ATPase activity was inhibited by increased sodium-pump inhibitors, NO release decrease may also play an important role in the pathogenesis of hypertension.  相似文献

AIM: To observe the myocardial protective effects of trimetazidine on myocardial infarction (MI) in Sprague-Dawley (SD) rats. METHODS: Ninety SD rats were randomly assigned to 3 groups (n=30 each): myocardial infarction group (MI group), MI+trimetazidine group (MT group) and sham group (S group). By permanently ligating the left anterior descending artery, the MI model was set up in the rats in MI group and MT group. Before and after setting up the MI model, normal saline was given to the rats in MI and S group by gavage. On the other hand, trimetazidine (3 mg/kg,twice per day) was given to the rats in MT group by gavage. At 8 h, 24 h and 48 h after applying trimetazidine, the serum level of cardiac troponin I (cTnI) was measured. At the 1st week, 2nd week and 4th week after treated with trimetazidine, the size of myocardial infarction, the maximum rising rate of the left ventricular systolic pressure (+dp/dtmax) and the maximum descending rate of the left ventricular diastolic pressure (-dp/dtmax) were measured. Also at the 1st week after applying trimetazidine, the cardiomyocyte apoptotic index was detected. RESULTS: Compared with MI group 2 weeks after applying trimetazidine, +dp/dtmax significantly increased in MT group , and -dp/dtmax also significantly increased in MT group . Four weeks after applying trimetazidine, +dp/dtmax significantly increased in MT group , and -dp/dtmax also significantly increased in MT group . At 8 h and 48 h after applying trimetazidine, no statistically significant difference (P>0.05) of serum cTnI between MI group and MT group was observed. However, at 24 h after applying trimetazidine, the serum level of cTnI decreased in MT group as compared with MI group . Aditionally, trimetazidine significantly decreased the infarction size of myocardium in MT group (0.248±0.052) as compared with MI group (0.362±0.082, P<0.01). CONCLUSION: Trimetazidine has short-term cardioprotective effects on the rats with acute MI by improving myocardial systolic and diastolic functions, reducing infarct size and inhibiting apoptosis.  相似文献

AIM: To evaluate the effect of rosuvastatin combined with irbesartan on the remodeling of myocardial hypertrophy in the rats. METHODS: Male SD rats(n=50) were randomly divided into control group, model group, rosuvastatin group, irbesartan group and combination group. The model of myocardial hypertrophy was established by subcutaneous injection of isoproterenol at dose of 2.5 mg/kg for 14 d. From the first day of modeling, the rats in control group and model group received intragastrical saline, and the rats in rosuvastatin group, irbesartan group and combination group were treated with rosuvastatin(4 mg·kg-1·d-1), irbesartan(15 mg·kg-1·d-1) and rosuvastatin(4 mg·kg-1·d-1)+ irbesartan(15 mg·kg-1·d-1), respectively. The interventions continued for 4 weeks. After the interventions, the cardiac mass index and left ventricular mass index of the SD rats were measured. Besides, the degree of myocardial hypertrophy was observed with HE staining. The mRNA expression of hypertrophy-related factors, such as ANF, β-MHC and AT1R was determined by RT-PCR, and the protein expression of AT1R was determined by Western blot. RESULTS: Compared with control group, the cardiac mass index, left ventricular mass index, as well as the mRNA expression of ANF and β-MHC in model group were significantly increased(P<0.05). Compared with model group, the above factors in rosuvastatin group and irbesartan group were decreased(P<0.05), and the factors in combination group were lower than those in rosuvastatin group and irbesartan group(P<0.05). In addition, the expression of AT1R at mRNA and protein levels in rosuvastatin group and irbesartan group was lower than that in model group(P<0.05), while the expression AT1R at mRNA and protein levels in combination group was lower than that in rosuvastatin group and irbesartan group(P<0.05). CONCLUSION: Rosuvastatin and irbesartan are equally effective drugs to resist the formation of myocardial hypertrophy by decreasing the expression of AT1R. Moreover, combination of the 2 drugs is more effective to improve the degree of myocardial hypertrophy than the 2 drugs alone.  相似文献

AIM: To investigate the protective effect of granulocyte colony-stimulating factor (G-CSF) combined with ischemic postconditioning (IP) on acute myocardial infarction (AMI). METHODS: Male New Zealand rabbits were randomly divided into 4 groups (n=15) after 30 min of left ventricular artery (LVA) occlusion: the rabbits in ischemia-reperfusion (IR) group were directly given reperfusion|the rabbits in G-CSF group were subsequently treated with G-CSF (10 μg·kg-1·d-1) by subcutaneous injection after direct reperfusion|the rabbits in IP group received 4 episodes of 30 s reperfusion and 30 s occlusion before total reperfusion|the rabbits in IP combined with G-CSF (IP+G-CSF) group were treated with both IP and G-CSF. Electrocardiogram (ECG) monitoring was performed during the operation. Blood was drawn to evaluate white blood cell count (WBC) and cardiac troponin I (cTnI) before operation and 7 d later. Ultrasound cardiography was performed to evaluate left ventricular remodeling and functions 4 weeks after operation. The sizes of infarcted myocardium were determined by triphenyltetrazolium chloride (TTC) staining. Apoptosis of cardiomyocytes was measured by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL) staining. RESULTS: ST-segment resolutions were significantly decreased in IP group and IP+G-CSF group compared with direct reperfusion groups (P<0.05). WBC significantly increased in the groups treated with G-CSF for 1 week. The values of cTnI after operation were significantly lowered in G-CSF group, IP group and IP+G-CSF group as compared with IR group (P<0.05). Left ventricular ejection fraction, the size of infarcted myocardium and apoptosis of cardiomyocytes were better in IP group, G-CSF group and IP+G-CSF group than those in IR group. CONCLUSION: G-CSF combined with IP is a promising strategy against cardiac reperfusion injury and accelerates cardiac repair in AMI.  相似文献

AIM: To verify the effect of cholecystokinin octapeptide(CCK-8) on cardiac function in endotoxin shock (ES) rats. METHODS: The rats were divided into four groups:control,lipopolysaccharide(LPS),CCK-8 and CCK-8+LPS. The left ventricle pressure(LVP),the maximal/minimum rate of LVP,heart rate (HR) and mean arterial pressure (MAP) were measured. The activity of superoxide dismutase (SOD),the contents of malondialdehyde (MDA) and nitric oxide (NO) in both serum and myocardium were also measured,respectively. RESULTS: CCK-8 (40 μg·kg-1, iv) elicited bradycardia in short time and gently increase MAP,LVP and ±LVdp/dtmax. Lipopolysaccharide(LPS, 8 mg·kg-1, iv) caused a variation in heart rate (HR)(a bradycardia following a tachycardia) and rapid decreases in MAP,LVP and ±LVdp/dtmax. The rapid variation of HR and the decline of MAP,LVP and ±LVdp/dtmax were reversed by pretreatment with CCK-8 in ES rats, but didn't restore to normal. The activity of SOD was increased and the contents of MDA and NO were decreased by pretreatment with CCK-8 in ES rats. CONCLUSION: The decline of cardiac function in ES rats could be reversed by pre-administration of CCK-8 and the decrease in NO production may be one of the mechanisms.  相似文献

AIM:To observe the expression of corticotropin releasing hormone (CRH) within the paraventricular nucleus of hypothalamus (PVN) and to explore the relationship between the activated CRH-containing neurons and sympathetic activity in rats with heart failure (HF).METHODS:Healthy male Sprague-Dawley (SD) rats were subjected to coronary artery ligation to induce HF,and chronic intracerebroventricular (ICV) infusion was performed by osmotic pump for 4 weeks.The rats in sham group and HF group were given vehicle (VEH;artificial cerebrospinal fluid 0.25 μL/h).The rats in HF plus treatment group were treated with CRH competitive inhibitor αh-CRH (15 mg/h).Meanwhile,the Lewis rats and Fischer 344 rats for control study also underwent coronary ligation to induce HF or sham surgery.After 4 weeks,left ventricular end-diastolic pressure (LVEDP) and maximum positive/negative change in pressure over time (±dp/dtmax) were determined.The right ventricular-to-body weight (RV/BW) and lung-to-body weight (lung/BW) ratios were calculated.The renal sympathetic nerve activity (RSNA) was recorded and the plasma norepinephrine (NE) level was measured.The expression of CRH in the PVN combined with the plasma adrenocorticotrophic hormone (ACTH) levels were measured.RESULTS:Compared with the sham-SD rats,the HF-SD rats had a greater number of CRH positive neurons in the PVN (accordingly the plasma ACTH levels were increased),accompanied by decreased±dp/dtmax and increased RSNA,plasma NE,LVEDP,lung/BW and RV/BW.However,ICV treatment with αh-CRH attenuated these changes in the HF-SD rats (P<0.05).Compared with the sham-Fisher 344 rats,the HF-Fisher 344 rats also had a greater number of CRH positive neurons in the PVN (accordingly the plasma ACTH levels were increased).In addition,they had significantly increased RSNA and plasma NE level,higher LVEDP,RV/BW and lung/BW,and lower±dp/dtmax(P<0.05).Compared with the SHAM-Lewis rats,the HF-Lewis rats had not significantly changed in the above parameters.CONCLUSION:In CHF,the CRH-containing neurons in PVN are activated,thus aggravating cardiac function by increasing sympathoexcitation.  相似文献

AIM: To investigate the effect of a treatment proposal, which consisted of tongxinluo, atorvastatin and aspirin, on adventitial inflammation of early atherosclerosis in rabbit carotid artery. METHODS: The atherosclerotic model was established in the rabbits with silicone collar, which was positioned around the carotid arterial adventitia+high-cholesterol diet. New Zealand rabbits (n=72) were randomly divided into 6 groups (n=12): control group, model group, tongxinluo group, atorvastatin group, aspirin group, and three-drug combination group. The rabbits in control group were fed with common foodstuffs, and the rabbits in all the other groups were fixed the right carotid arteries with the silicone tube, and were fed with fatty foodstuffs. The rabbits in tongxinluo group, atorvastatin group and aspirin group were given the suspension of tongxinluo supermicropowder (0.3 g·kg-1·d-1), atorvastatin (2.5 mg·kg-1·d-1) and aspirin (12 mg·kg-1·d-1) respectively,and the rabbits in three-drug combination group were given the suspension of tongxinluo supermicropowder (0.3 g·kg-1·d-1), atorvastatin (2.5 mg·kg-1·d-1) and aspirin (12 mg·kg-1·d-1) together. The rabbits in each group were fed with the corresponding medicines for 4 weeks. The tissue slices of carotid artery were observed under light microscope with HE staining. The change of blood lipid was detected by biochemical assay. The protein levels of MCP-1, IL-1β and IL-10 in the carotid arterial adventitia were detected by ELISA. The immunohistochemical staining was used to detect the protein expression of IL-8 around the carotid arterial adventitia.RESULTS: Compared with control group, the contents of TC, TG and LDL-C were significantly increased, and the content of IL-10 was significantly decreased in model group. The levels of TC, TG and LDL-C were significantly decreased in tongxinluo group and atorvastatin group compared with model group, no significant difference between tongxinluo group and atorvastatin group was observed. In the three-drug combination group, the levels of TC, TG and LDL-C were lower than those in atorvastatin group and tongxinluo group. Compared with control group, the contents of MCP-1 and IL-1β were significantly increased, and the content of IL-10 was significantly decreased in model group. Compared with model group, the contents of MCP-1 and IL-1β were decreased in tongxinluo group, atorvastatin group and aspirin group, no significant difference between the 3 groups was observed. The content of IL-10 was decreased in three-drug combination group, and the contents of TC, TG and LDL-C were lower than those in tongxinluo group, atorvastatin group and aspirin group. The content of IL-8 was decreased in tongxinluo group, atorvastatin group, aspirin group and three-drug combination group.CONCLUSION: The strategy of three-drug combination enhances the effect of regulating the lipid metabolism and inhibiting the adventitia inflammation. It plays an important role to intervene in the process of atherosclerosis.  相似文献

AIM: To investigate the changes of matrix metalloproteinases (MMP2 and MMP9) and tissue inhibitor of metalloproteinases (TIMP2 and TIMP1) in the myocardial fibrosis induced by cyclosporin A (CsA), and to explore the underlying mechanism. METHODS: Female Wistar rats (n=64, 6~8 weeks old) weighing (200±25) g were randomly divided into 4 groups[control, low dose of CsA (5 mg·kg-1·d-1), medium dose of CsA (12.5 mg·kg-1·d-1) and high dose of CsA (25 mg·kg-1·d-1)]. The rats were intraperitoneally injected with saline and different doses of CsA, respectively. CsA was continuously administered for 1, 2 and 3 weeks, and then the animals were killed to collect samples. HE staining was used to observe the morphological structure of myocardium. Masson staining was used to observe the deposition of myocardial collagen and the degree of myocardial fibrosis. The protein expression levels of MMP2, MMP9, TIMP2 and TIMP1 were determined by immunohistochemistry and Western blot. RESULTS: HE staining showed that CsA induced cardiomyocyte edema, eosinophilic changes in the cytoplasm, fine granular and vacuolar changes in cardiomyocytes, disappearance of myocardial striae, nuclear condensation, and myocardial focal necrosis and fibrosis. Masson staining showed that the degree of myocardial interstitial fibrosis was getting worse with the increase in the duration and doses of CsA exposure. The results of immunohistochemistry and Western blot showed that the expression of MMP2 in CsA groups was significantly increased in the process of CsA-induced myocardial fibrosis. In each CsA dose group, MMP2 was highly expressed at the first week, and the expression was gradually decreased over time. In contrast, the expression of TIMP2 was increased gradually in a time-and dose-dependent manner (P<0.05). Compared with control group, MMP9 expression level was low at the first week, increased at the second week, and decreased at the third week (P<0.05). Compared with control group, the expression of TIMP1 was dose-dependently increased (P<0.05). CONCLUSION: CsA induces myocardial injury and interstitial fibrosis in rats in a dose-and time-dependent manner. CsA-induced myocardial fibrosis is associated with changes of MMPs and TIMPs, and is affected by the imbalance of MMPs/TIMPs.  相似文献

AIM: To determine the effects of Tongxinluo(TXL) on connexin 43(Cx43) remodeling and ventricular arrhythmia(VA) after myocardial infarction(MI) in rats. METHODS: Male SD rats were randomly divided into sham-operated(sham) group(n=25) and operation group(n=75). The left anterior descending(LAD) was ligated in operated group, while the rats in sham group only underwent pericardiotomy. The rats in operation group which survived for 3 d after operation were randomly assigned to TXL group and MI group. The rats in TXL group was administrated with TXL(2 g·kg-1·d-1, intragastric administration) for 4 weeks, while normal saline was applied to the rats in sham group and MI group. The levels of interleukin-1β(IL-1β) and endothelin-1(ET-1) in the tissue from the border zone were measured by ELISA after treatment. The distribution and the mRNA and protein expression of Cx43 were detected by immunohistochemical staining, RT-PCR and Western blotting, respectively. The burst pacing was used to induce ventricular arrhythmia(VA). RESULTS: Compared with sham group, the levels of IL-1β and ET-1 and the incidence of VA were significantly increased, while the mRNA and protein expression of Cx43 was markedly reduced with irregular distribution in MI group(P<0.05). Compared with MI group, the levels of IL-1β and ET-1 and the incidence of VA were significantly reduced, while the expression of Cx43 at mRNA and protein levels was markedly increased with augmented linear distribution in the myocardial cell intercalated disc in TXL group(P<0.05). CONCLUSION: TXL reduces the incidence of VA after MI via inhibiting the Cx43 remodeling.  相似文献
