AIM: To investigate the role of fatty acid translocase/CD36 (FAT/CD36) in adipose tissue inflammation induced by a high-fat diet. METHODS: C57BL/6J mice were fed with a normal-chow diet (NCD) or a high-fat diet (HFD) for 14 weeks. The content of free fatty acid (FFA) in the serum was measured by ELISA. The expression of CD36, cytokines and chemokines at mRNA and protein levels in the adipose tissues was determined by real-time polymerase chain reaction and Western blotting. Immunohistochemical staining was used to examine the macrophages infiltration in the adipose tissues. The inflammatory responses in CD36 knockout mice and wild type mice with high-fat diet were analyzed. RESULTS: The levels of FAT/CD36 were higher in HFD group than that in NCD group. HFD feeding enhanced the mRNA and protein expression of IL-1β, IL-6, TNF-α, MCP-1 and MIP-1, as well as promoted macrophage infiltration in the adipose tissues. Interestingly, as fed with HFD, the expression of cytokines/chemokines and macrophage infiltration were significantly reduced in adipose tissues of the CD36 knockout mice, compared with the wild type mice. CONCLUSION: High-fat diet promotes adipose tissue inflammation in the mice in a FAT/CD36-dependent manner.  相似文献

AIM: To investigate whether the protective effect of adiponectin on glucose and lipid metabolism is achieved through down-regulating major histocompatibility complex class Ⅱ (MHCⅡ) in the adipose tissue. METHODS: Adiponectin knockout (KO) mice and C57BL/6(WT) mice were fed with high-fat diet and standard diet for 24 weeks, respectively. The body weight, fasting blood glucose (FBG), fasting insulin (FINS), homeostasis model assessment of insulin resistance (HOMA-IR), triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), hepatic histology, and class Ⅱ trans-activator (CⅡTA), histocompatibility 2 class Ⅱ antigen E beta (H2-Eb1) and cluster of differentiation 74(CD74) mRNA and MHC Ⅱ protein levels in adipose tissue were measured at sacrifice. siRNA targeting MHC Ⅱ and overexpression vector was used in 3T3-L1 cells to explore the effect of adiponectin on the protein level of MHCⅡ. RESULTS: The levels of body weight, FBG, FINS, HOMA-IR, TC, TG, LDL-C, hepatic steatosis, CⅡTA, H2-Eb1 and CD74 mRNA expression, and MHCⅡ protein expression in the KO mice were higher than those in the WT mice that fed with high-fat diet or standard diet. In 3T3-L1 cells, inhibition of adiponectin reversed MHC Ⅱ protein level induced by specific siRNA. The expression of MHC Ⅱ in adipocytes decreased after adiponectin was overexpressed. CONCLUSION: Adiponectin improves glucose and lipid metabolism through suppressing the expression of MHCⅡ in the adipose tissue.  相似文献

AIM To investigate the effects of cytochrome P450 (CYP450) epoxygenase/epoxyeicosatrienoic acid (EET) pathway on insulin resistance in obese mice, and to explore the possible mechanisms. METHODS High-fat diet-induced obesity model was established in C57BL/6Cnc mice, and the obese mice were randomly divided into 3 groups, including obesity group (treated with saline; n=10), EET group (treated with 11,12-EET; n=10) and EET inhibitor 14,15-epoxyeicosa-5(Z)-enoic acid (EEZE) group (n=10). Normal C57BL/6Cnc mice (n=10) treated with saline served as control. Protein expression of CYP2J2 (one of CYP450 epoxygenases) and hypoxia-inducible factor-1α (HIF-1α) was measured by Western blot. Vessel-like structure was detected by immunofluorescence staining. The serum levels of insulin, tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and monocyte chemoattractant protein-1 (MCP-1) were measured by ELISA. RESULTS In obese mice, homeostasis model assessment of insulin resistance (HOMA-IR) values were increased, the protein level of CYP2J2 was reduced, and the protein level of HIF-1α was increased in adipose tissues as compared with the controls (P<0.05). The serum levels of MCP-1, IL-1β, IL-6 and TNF-α were also significantly increased in obese mice (P<0.05). After treatment with 11, 12-EET, the HOMA-IR values were decreased compared with vehicle-treated obese mice, HIF-1α expression levels were decreased in the adipose tissue, and the serum levels of MCP-1, IL-1β, IL-6 and TNF-α were reduced (P<0.05). Immunohistochemical results of adipose tissue from vehicle-treated obese mice showed a marked decrease in vessel-like structures (CD31-positive) compared with normal control mice (P<0.05). EET treatment significantly increased the newly formed vessel-like structures in the visceral adipose tissues of obese mice as compared with vehicle-treated obese mice (P<0.05). CONCLUSION High-fat diet-induced obesity and insulin resistance are closely related to the CYP450 pathway. Exogenous EETs effectively decrease obesity-induced insulin resistance possibly through pro-angiogenesis and attenuation of hypoxia and inflammation.  相似文献

AIM:To observe the relationship between UCP2 mRNA expression in white adipose tissue and diet-induced obesity in SR-A I/II gene knock-out (SR-AⅠ/Ⅱ-/-) mice. METHODS:Fluorescent quantitative RT-PCR was used to detect UCP2 mRNA expression in mice epididymal white adipose tissue. The cellular morphological changes were analyzed by using image analysis. Serum TG, TC and LDL-C concentrations were measured by enzymatic determination. RESULTS:After fed with high fat diet for 12 weeks, average body weight of SR-A I/II-/- mice was much higher than that of wild type (SR-A I/II+/+) control mice (P<0.01), as well as the serum lipid (TG, TC, LDL-C) levels, epididymal fat pad weight, adipocyte area and diameter. UCP2 mRNA expression in white adipose tissue in SR-A I/II-/- mice was significant lower than that in SR-A I/II+/+ control mice (P<0.01). CONCLUSION:Abnormal UCP2 mRNA expression may be associated with SR-A I/II-/- mice susceptive to diet-induced obesity.  相似文献

AIM: To investigate the effects of glucagon-like peptide-1(GLP-1) receptor agonist exendin-4 on white adipose tissue (WAT) and the underlying mechanisms. METHODS: Male C57BL/6J mice (8 weeks) were challenged by high-fat diet for 12 weeks, and were randomly divided into saline group and exendin-4 group. The mRNA expression of sirtuin 1(SIRT1), adipose triglyceride lipase (ATGL), TNF-α and adiponectin of WAT was detected by real-time PCR. 3T3-L1 adipocytes or mouse embryonic fibroblasts cells were treated with exendin-4 for 24 h. The protein levels of SIRT1, ATGL and hormone-sensitive lipase (HSL) were determined by Western blot.RESULTS: Exendin-4 significantly decreased epididymal fat weight, fasting blood glucose and serum triglyceride levels (P<0.05), and reduced body weight and serum TNF-α level. The mRNA expression of SIRT1, ATGL and adiponectin in WAT was all significantly up-regulated by exendin-4, which were contrary to the down-regulation of TNF-α mRNA expression (P<0.05). Exendin-4 promoted the protein expression of SIRT1, ATGL, and HSL in 3T3-L1 adipocytes in a dose-dependent manner. Less lipid droplets with up-regulation of lipolytic protein expression were observed when combined with SIRT1 agonist treatment, which were suppressed by SIRT1 inhibitor. Deletion of SIRT1 led to larger adipocytes with more lipid droplets, and the effect of exendin-4 on the lipolysis disappeared when SIRT1 was deficient.CONCLUSION: Exendin-4 promotes lipolysis in WAT of obese mice via activation of SIRT1.  相似文献

ATM: To investigate the effect of oxymatrine (OXY) on high fat-induced insulin resistance in mice, and to investigate the mechanism. METHODS: ApoE-/-mice with high-fat diet for 16 weeks were divided into insulin resistance group, and OXY groups at concentrations of 25, 50 and 100 mg/kg. C57BL/6J mice served as normal control group. The mice in OXY groups were gavaged with OXY for 8 weeks. Glucose tolerance test in the mice was performed. Fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), fatty acid (FFA) and fasting insulin (FINS) in the plasma were measured. The mRNA expression of insulin receptor (INSR), insulin receptor substrate-2 (IRS-2), glucose transporter 2 (GLUT2) in the liver tissues was examined by RT-qPCR. The protein levels of GLUT2, INSR, IRS-2, p-INSR, p-IRS-2, PI3K, p-PI3K, serine/threonine protein kinase (AKT) and p-AKT were examined by Western blot.RESULTS: OXY reduced the levels of FBG, TC, TG, FFA and FINS, and attenuated insulin resistance. Compared with insulin resistance group, the mRNA expression of INSR, IRS-2 and GLUT2 significantly increased in OXY groups (P<0.05). The protein levels of p-INSR/INSR, p-IRS-2/IRS-2, p-PI3K/PI3K, p-AKT/AKT and GLUT2 also increased in OXY groups (P<0.05).CONCLUSION: OXY ameliorates high fat-induced insulin resistance in mice via PI3K/AKT pathway.  相似文献

AIM: To study whether the pulmonary infection of Escherichia coli (E. coli) interferes the glucolipid metabolism in high-fat diet-induced obese mice. METHODS: High-fat diet-induced obese mice (n=48) and normal chow-fed control mice (n=48) were intranasally infused with 40 μL fluid containing 4×109 CFUs E. coli. The serum, periepididymal adipose tissue and liver were obtained at 0 d, 1 d, 2 d, 3 d and 4 d after infection. The body mass, periepididymal adipose tissue and liver were weighed, and the levels of fasting blood glucose (FBG), fasting blood insulin (FINS), free fatty acid (FFA) and very-low-density lipoprotein (VLDL) were measured by ELISA. The serum total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C), and hepatic TG contents were detected, and the hepatic steatosis was observed under microscope with oil red O staining. RESULTS: Compared with day 0, the body mass, fat mass and fat index were decreased significantly from day 1 to day 4 after infection (P<0.05). The levels of FBG, FINS and HOMA-IR were apparently raised from day 2 to day 4 after infection (P<0.05). The contents of serum FFA, TG and VLDL were increased markedly from day 1 to day 4 after infection (P<0.05). However, the concentrations of serum TC, LDL-C and HDL-C were decreased obviously from day 1 to day 3 (P<0.05). The liver mass, liver index and TG content were significantly increased from day 1 to day 4 (P<0.05). Consistently, the lipid droplet accumulation in the liver cells was increased obviously at day 2 and day 4 after infection. Compared with control group, except the levels of serum TC, LDL-C and HDL-C in obese group substantially decreased, the other indexes were increased by different degrees during the whole experiment period (P<0.05). CONCLUSION: Pulmonary infection of Escherichia coli exacerbates the disorder of glucose and lipid metabolism in high-fat diet-induced obese mice, which contributes the development of insulin resistance and hepatic steatosis.  相似文献

AIM: To study the effects of Shan He Jian Fei Granule (SHJFG), a Chinese medicine, on weight-reduction and fat-decrease in adiposity rats, and to observe the changes of leptin, adiponectin and resistin. METHODS: After the models were prepared successfully, the rats were randomly divided into four groups: normal group, low dosage, middle dosage, high dosage and obesity control group. After 8 weeks interference with SHJFG, the weight and the naso-anal length of each rat was measured and Lees index were calculated. The levels of TG, TC, LDL, HDL and leptin in serum were carefully determined. The gene expressions of adiponectin and resistin in adipose tissues of rats were determined by RT-PCR. RESULTS: Compared to high-fat diet group, the body weights, the Lees indexes, the weight of fat tissues and levels of TG, TC, LDL and leptin in SHJFG groups significantly decreased (P<0.05). Adiponectin mRNA expression in SHJFG group significantly increased (P<0.05), and the resistin mRNA expression also significantly increased. CONCLUSION: SHJFG significantly decreases the body weight and the serum levels of TG, TC and LDL in obese rats. The effects of SHJFG in raising the mRNA expressions of adiponectin and resistin in fat tissues may be one of the main role that results in lowering body weight in obese rats.  相似文献

AIM: To observe the pathologic changes of liver in diabetic rats and to investigate the role of mRNA expression of insulin receptor and leptin receptor in the pathogenesis of nonalcoholic fatty liver disease (NAFLD). METHODS: Twenty male Sprague-Dawley rats were divided randomly into two groups: normal control group and diabetic group. After fed with high-fat diet for 4 weeks, diabetic rats were injected with streptozotocin at a dosage of 30 mg/kg intraperitoneally to induce NAFLD model of type 2 diabetes mellitus. Then the diabetic animals were fed with high-fat diet continuously for 12 weeks. At the end of the experiment, the rats were sacrificed, the concentrations of blood glucose, serum lipid, ALT and AST were measured biochemically. The levels of serum leptin and serum insulin were detected by enzyme-linked immunosorbent assay (ELISA) and radio immunoassay (RIA), respectively. The pathologic changes of liver were observed under light microscopy (LM) stained with HE, Sudan Ⅲ and Masson trichrome staining, respectively. The ultra-structural changes of liver were observed under transmission electron microscopy (TEM). Additionally, the mRNA expressions of PEPCK, G6Pase, insulin R and leptin R from rat livers were assayed by semi-quantitative RT-PCR. RESULTS: The levels of blood glucose, serum insulin, serum TG, ALT and AST increased significantly (P<0.01), serum TC elevated (P<0.05), and the levels of serum leptin decreased (P<0.01) in diabetic group compared to those in normal control group. Obvious liver fatty degeneration, piecemeal necrosis with accompanying inflammatory infiltration and fibrosis were found under LM. Hepatocytes pyknosis, lots of lipid deposits in cytoplasm of hepatocytes, proliferation of collagen in space of Disse were observed under TEM in diabetic group. The expression of insulin R and leptin R mRNA in liver from diabetic rats increased significantly (P<0.01) while the expression of PEPCK and G6Pase mRNA remained unchanged. CONCLUSION: Insulin resistance plays an important role in the pathogenesis of NAFLD. Low level of serum leptin, up-regulation of mRNA expression of insulin R and leptin R in liver caused by insulin resistance may be involved in this process.  相似文献

AIM: To investigate the effect and mechanism of Astragalus polysaccharide (APS) on the amelioration of hepatic insulin resistance in high fat-fed mouse model.METHODS: C57BL/6J mice (n=26) were divided into three groups randomly: C group (an animal model for control,n=10);IR group ( an animal model of insulin resistance,n=8) and IA group (an animal model in high-fat diet with APS treatment for12 weeks,700mg·kg-1·d-1,ig).High-fat diet was used to induce the formation of insulin resistant.The parameters and insulin sensitivity of the animals were observed.The pathological features of the liver were presented through microscope and TEM.The expression changes of hepatic GSK3β were measured by Western blotting.RESULTS: In this study,the fat-fed mouse model of insulin resistance was established successfully.The mice in IA group responded to the 12-week APS therapy with a significant decrease in the level of blood glucose,plasma insulin,body weight,hepatic TG/FFA and improved glucose tolerance compared with those in IR group.In addition,the expression and the activity of GSK3β were lower in IA group (vs IR group,P<0.05).We also found the hepatic steatosis could be significantly alleviated with APS therapy.CONCLUSION: These results indicate that APS prevents the occurrence of insulin resistance and the hepatic steatosis induced by high-fat diet,at least in part by inhibiting the expression and activity of the hepatic GSK3β.  相似文献

AIM: To observe the effects of losartan potassium on renal expression of transforming growth factor beta 1(TGF-β1), CD68 and monocyte chemoattractant protein-1 (MCP-1) in type 2 diabetic nephropathy rats for exploring the protective mechanism of losartan potassium on type 2 diabetic rat kidney. METHODS: Thirty Sprague-Dawley rats were randomized into 3 groups: normal control group, model group and treatment group. The morphology of kidney tissues, the renal function, and the change of 24 h urinary protein quantitative index were measured after 15 weeks of treatment, while TGF-β1, CD68 and MCP-1 expression in kidney cortex was observed by immunohistochemistry. RESULTS: Compared with the normal control rats, the body weight of the rats was lower in other groups, but the levels of blood glucose, triglyceride and cholesterol were higher.The expression of CD68, MCP-1 and TGF-β1, 24 h urinary protein quantitative index and serum creatinine were higher in model group than those in normal control rats. However, compared with model group, serum creatinine, 24 h urinary protein quantitative index and the expression of CD68, MCP-1 and TGF-β1 were decreased in treatment group. CONCLUSION: Losartan potassium protects the kidney of diabetic nephropathy rats through inhibiting the expression of TGF-β1 and MCP-1 in the kidney and restraining macrophage infiltration.  相似文献

AIM:To explore the effect of L-glutamine (Gln) on obesity and insulin resistance in high-fat diet(HFD)-induced C57BL/6J mice. METHODS:Male C57BL/6J mice (n=60) were randomly divided into normal control (NC) group, HFD group, HFD+L-alanine (Ala) group and HFD+Gln group. Each group had 15 mice. The body weight of the mice was recorded weekly. Fasting blood glucose (FBG) of the mice was tested after 12-h fasting only with water-drinking at the end of the 16th week. The mice were sacrificed and epididymal fat pad was measured. The levels of insulin (INS), leptin (LEP), adiponectin (APN) and glucagon-like peptide-1 (GLP-1) were measured by ELISA. The insulin resistance index (IRI) and insulin sensitivity index (ISI) were calculated. RESULTS:Compared with NC group, the body weight, epididymal fat pad weight, and the levels of FBG, INS, IRI and LEP increased significantly in HFD group (P<0.05), while the levels of ISI and APN decreased significantly (P<0.05). Compared with HFD group, the body weight, and the levels of FBG, IRI and LEP decreased significantly in HFD+Gln group (P<0.05), while the levels of ISI and APN increased significantly (P<0.05). No significant difference of serum GLP-1 levels the four groups was observed. CONCLUSION:L-glutamine reduces the body weight and attenuates the insulin resistance of HFD-induced mice.  相似文献

AIM: To explore the regulatory mechanism of LPIN1 in hepatic insulin resistance by investigating the influence of metformin on the expression of LPIN1 and AMP-activated protein kinase(AMPK) signaling in the rats with high-fat diet-induced insulin resistance. METHODS: Thirty-six 4-week-old male Wistar rats were randomly divided into 2 groups: control group and high-fat diet (HF) group. The rats in HF group were fed with high-fat diet for 8 weeks and then were randomly divided into 2 subgroups: HF group and metformin intervention group, and the animals were continuously raised for 8 months. The mRNA levels of α1 and α2 subunit of AMPK as well as LPIN1 were measured by real-time RT-PCR. Phospho-AMPKα (Thr-172) was detected by Western blotting to evaluate the activity of AMPK. RESULTS: After 4 months, the rats in HF group showed significant increase in the levels of body weight, fast plasma glucose and insulin, and the levels of triglyceride and total cholesterol significantly elevated.Significant decrease in LPIN1 and phospho-AMPKα (Thr-172) expression in the rat livers were also observed. After treated with metformin, the metabolic indexes of the HF rats were improved. The mRNA and protein expression of AMPKα1 and AMPKα2 had no significant difference among the 3 groups. Metformin treatment also increased the expression of LPIN1 in the liver tissues of HF rats. CONCLUSION: The decrease in LPIN1 expression and AMPK activity may contribute to hepatic insulin resistance in diet-induced obese rats. Metformin improves the LPIN1 expression and AMPK activity through the interaction between LPIN1 and AMPK signal pathways.  相似文献

AIM: To investigate the effect of electroacupuncture (EA) in different intensities on unfolded protein response (UPR) signaling pathway in adipose tissues from high-fat diet-induced obese rats. METHODS: Sixty SD rats were randomly divided into 2 groups: common diet group (n=10) and high-fat diet group (n=50). Thirty obese rats in high-fat diet group were chosen and divided into high-fat diet group, 5 mA EA group and 1 mA EA group (n=10 each). EA at frequency of 20 Hz and intensities of 5 mA or 1 mA was applied to ST 36 and SP 6 for 15 min once daily for 2 weeks. The body weight was detected once a week. The expression of p-PERK and CHOP/GADD153 in epididymal adipose tissues was determined by Western blotting. The content of Bcl-2 and caspase-12 in the adipose tissues was measured by ELISA. RESULTS: After EA, the body weight and the expression of p-PERK and CHOP/GADD153 in obese rats were significantly decreased (P<0.01 or P<0.05). The rats in 5 mA EA group exhibited a better improvement on the protein expression than the rats in 1 mA EA group.CONCLUSION: EA has a beneficial regulatory effect on the rats with simple obesity. Moreover, the EA density of 5 mA is superior to 1 mA.  相似文献

AIM: To study the effect and mechanism of extract of ginkgo biloba (EGB) on liver glucocorticoid receptor (GR) expression in type 2 diabetic rats. METHODS: Thirty male Sprague-Dawley rats were divided randomly into three groups: normal control group (n=10), type 2 diabetic group (n=10) and ginkgo biloba treated group (n=10). After fed with high-fat feeding for 4 weeks, the later two groups were injected with streptozotocin at a dose of 30 mg/kg intraperitoneally to induce type 2 diabetic rat model. The EGB treated group was gavaged with EGB at the dose of 50 mg·kg-1·d-1 for 12 weeks. At the end of experiment, the rats were sacrificed, the blood glucose, serum lipid and blood insulin were measured. The morphology of liver tissue was observed under light microscopy with HE staining. GR mRNA expression in liver was measured by RT-PCR. The level of GR protein expression in liver tissue was detected by immunohistochemistry. RESULTS: EGB reduced the levels of blood glucose, blood lipids, blood insulin in diabetic rats. EGB also relieved fatty degeneration and necrosis of the hepatic cells, ameliorated infiltration of inflammatory cells in the liver; and decreased GR expression at both mRNA and protein levels in diabetic liver. CONCLUSION: EGB may inhibit GR expression in liver of type 2 diabetic rats, which results in decreasing the level of blood glucose, blood lipid, blood insulin and relieving the liver damage in type diabetic rats.  相似文献

AIM: To study the role of Radix Pseudostellariae polysaccharide (RPP) in hepatic insulin resistance.METHODS: Six-week-old C57BL/6J mice were randomly divided into low-fat diet (LFD) control group and high-fat diet (HFD) model group. After 16 weeks, intraperitoneal pyruvate tolerance test (IPPTT) was performed to determine the establishment of the HFD-induced hepatic insulin resistance model. HFD containing RPP (500 mg/kg) was given for 4 consecutive weeks. IPPTT, liver malondialdehyde (MDA) level and liver mitochondrial MDA level were measured. The protein levels of p-AKT (Ser473/Thr308), p-AMPK, nuclear factor E2-related factor 2 (Nrf2), NQO1 and IκBα in the liver tissues were measured by Western blot.RESULTS: After administration of RPP, a significant reduction in the levels of blood glucose and hepatic mitochondrial MDA was observed. The levels of p-AKT (Ser473/Thr308) and p-AMPK were significantly elevated in the liver tissues. The hepatic IκBα levels were up-regulated. RPP also enhanced the expression of Nrf2 system-regulated proteins NQO1 and HO-1 in the liver tissues.CONCLUSION: Radix Pseudostellariae polysaccharides effectively reduce HFD-induced hepatic insulin resistance in C57BL/6J mice and improves liver glucose metabolism by ameliorating HFD-impaired hepatic transduction of insulin signaling, activating Nrf2-associated signaling and inhibiting the expression of inflammatory signaling proteins.  相似文献
