A homothallic Phytophthora species was found to be consistently associated with a rot of mature fruits of two local cultivars of olive (Olea europaea) in Calabria, southern Italy. The phylogenetic analysis of sequences of the ITS1‐5.8S‐ITS2 region and cox1 gene enabled its identification as a new species of clade 2, with a basal position compared to previously described subclades. The new species is described formally with the epithet Phytophthora oleae, referring to the natural matrix from which it was isolated. A unique combination of molecular and morphological characters clearly separates P. oleae from other already described Phytophthora species. This new species produced semipapillate, occasionally bipapillate, persistent sporangia on simple sympodially branching sporangiophores as well as globose and smooth‐walled oogonia, paragynous antheridia and spherical, plerotic oospores. The pathogenicity of P. oleae was confirmed in inoculation trials on fruits of three olive cultivars, including the two local cultivars from which the pathogen had been isolated.  相似文献

Chinese rose (Rosa chinensis) is one of the most popular and widely cultivated flowers worldwide and has extremely high economic and ornamental value. In 2020 wilt disease on R. chinensis was discovered in Pukou District, Nanjing, Jiangsu Province, China. Fungal isolates were obtained from the stems of the rose. According to morphological characteristics and multilocus phylogenetic analyses with the sequences of the rDNA internal transcribed spacer (ITS), translation elongation factor 1-α gene (TEF1-α), and part of the RNA polymerase II gene (RPB2), the isolates YJ1 to YJ4 were determined as a new species of Fusarium solani species complex, and named as Fusarium rosicola sp. nov., which is hereby described and illustrated. Pathogenicity of the isolate YJ1 was verified by Koch's postulates. The fungus was determined as the pathogen causing rose vascular wilt. The isolate YJ1 was labelled with green fluorescent protein (GFP), and roots of R. chinensis were inoculated. The result showed that the fungus infected the vascular tissue of the host plants and caused withering of the above-ground parts, resulting in the death of the whole plant. The GFP-labelled pathogen was reisolated from the stems and foliage, proving that this is a newly emerged systemic disease on R. chinensis in the world.  相似文献

Isolates of an unknown Phytophthora species from the ‘Phytophthora citricola complex’ have been found associated with mortality of Aucuba japonica in the UK. Based on morphological characteristics, growth–temperature relationships, sequences of five DNA regions and pathogenicity assays, the proposed novel species is described as Phytophthora pachypleura. Being homothallic with paragynous antheridia and semipapillate sporangia, P. pachypleura resembles other species in the ‘P. citricola complex’ but can be discriminated by its distinctively thick‐walled oospores with an oospore wall index of 0·71. In the phylogenetic analysis based on three nuclear (ITS, β‐tubulin, EF‐1α) and two mitochondrial (cox1, nadh1) DNA regions, P. pachypleura formed a distinct clade within the ‘P. citricola complex’ with P. citricola s. str., P. citricola E and P. acerina as its closest relatives. Phytophthora pachypleura is more aggressive to A. japonica than P. plurivora and P. multivora and has the potential to affect other ornamental species.  相似文献

马铃薯干腐病是马铃薯最重要的贮藏期病害之一,现已成为马铃薯贮藏期烂窖的重要原因。实现病原菌的快速检测对病害诊断和科学防控具有重要的实践意义。本研究基于镰刀菌翻译延伸因子序列设计了一对检测接骨木镰刀菌Fusarium sambucinum的特异引物Fs-F/Fs-R。该特异引物可从接骨木镰刀菌中获得309bp的特异性扩增片段,而其他种类镰刀菌及马铃薯重要病害病原菌中均无此片段,说明该引物具有专一性。该体系的灵敏度检测结果表明,最低能检测到的接骨木镰刀菌基因组DNA浓度为70pg/μL。该引物也适用于发病马铃薯块茎中接骨木镰刀菌的快速检测。  相似文献

Basal rot is a common disease in lettuce greenhouses. A 3-year study on the diversity of pathogens associated with basal rot in Belgium was carried out. A total of 150 isolates were collected originating from 56 greenhouses. Four pathogens appeared to be involved. Rhizoctonia solani was found to be the causal agent at 23 locations, Sclerotinia spp. at 14, Botrytis cinerea at 17 and Pythium spp. at seven. The isolates of R. solani were further characterised to anastomosis groups and subgroups using morphological characteristics, pectic zymogram and PCR-RFLP. Five anastomosis groups could be distinguished: AG1-1B, AG4 HGI, AG10, AG2-1, AG2-1 Nt and AG3, with isolates of AG4 HGI and AG1-1B being the most prevalent and the most aggressive. Sclerotinia sclerotiorum was found at 13 locations, while S. minor was found at only one location. Based on ITS-sequencing Pythium isolates were assigned to three different species. At 20°C, isolates of all pathogens were able to cause lesions on detached lettuce leaves, except isolates of R. solani AG3 and AG2-1 Nt. A correlation could be found between the occurrence of the pathogens and the growing season. Botrytis cinerea was the most common pathogen in winter, whereas R. solani was most frequently isolated in summer. Sclerotinia spp. and Pythium spp. were isolated in spring, summer and autumn. The information obtained in this study will be most useful in the development of an alternative control strategy for causal agents of basal rot.  相似文献

A 3-year survey was undertaken to establish the relative frequency of different Fusarium spp. present as inoculum on potato tubers collected from four regions of Great Britain. A total of 219 samples (comprising 10 950 tubers) were collected from the 2000, 2001 and 2002 crops and processed to recover dry rot-producing isolates. In total, 228 isolates of Fusarium spp. were recovered. Most (94·7%) of these isolates were attributed to one of four Fusarium species: F. coeruleum, F. avenaceum, F. culmorum and F. sambucinum (formerly F. sulphureum) . The incidence of the combined Fusarium spp. increased the further south the crops had been grown. Fusarium coeruleum was the most commonly isolated species in each survey year, comprising 37 to 52% of the total Fusarium species. Selected isolates of each species were evaluated for their ability to produce rots in potato tubers. Fusarium sambucinum was a more aggressive pathogen than the other Fusarium species in eight out of 10 cultivars. Fusarium avenaceum and F. culmorum were relatively weaker pathogens. However, these species were aggressive on some cultivars, notably Hermes. The selected isolates were also assessed for their sensitivity to the fungicides thiabendazole and imazalil. Using in vitro tests, 65% of F. sambucinum isolates were resistant to thiabendazole and 7% of F. avenaceum isolates were resistant to imazalil. Tubers treated with imazalil yielded a higher proportion of isolates of F. avenaceum than those that were untreated. Similarly, a higher proportion of F. sambucinum isolates were recovered from tubers treated with thiabendazole than from those that were not treated.  相似文献

为探究石榴干腐病的病原菌种类及嘧菌酯对石榴干腐病菌的生物学活性,采用单孢分离法对石榴病果中的病原菌进行分离纯化,然后进行分子鉴定和致病性测定,并通过室内生测法测定了嘧菌酯对其菌丝生长和孢子萌发的影响,且连续2年开展了大田防治试验。经形态学、分子生物学鉴定及致病性测定结果表明,石榴干腐病的病原菌为石榴壳座月孢Pilidiella granati。嘧菌酯在水杨肟酸(SHAM)的协同作用下,对石榴干腐病菌的菌丝生长和孢子萌发具有强烈的抑制作用,对其菌丝生长和孢子萌发的平均EC50分别为0.202μg/m L和0.007μg/m L(含100μg/m L SHAM);在大田防治试验中,嘧菌酯对石榴干腐病具有良好的防效,其中1 000倍25%嘧菌酯SC稀释液在2013和2014年的防效分别可达90.85%和81.91%,显著高于其它处理。表明嘧菌酯可作为防治石榴干腐病的候选药剂。  相似文献

为明确河北和内蒙古马铃薯干腐病菌种类和优势种群,从两省采集300块具干腐病症状的样品,通过分离培养、形态鉴定、TEF-1α序列分析及致病性测定等,对马铃薯干腐病菌种类进行了系统研究.结果表明,两省马铃薯干腐病共存在4种病原菌,即接骨木镰刀菌Fusarium sambucinum、锐顶镰刀菌F.acuminatum、尖孢镰刀菌F.oxysporum和芬芳镰刀菌F.redolens,分别为149、80、3、3株.其中,河北省的优势种群为接骨木镰刀菌和锐顶镰刀菌,其发生频率分别为50%和48.68%;内蒙古的优势种群为接骨木镰刀菌,发生频率为87.95%.  相似文献

A new disease of rose was noticed in Chiba Prefecture of Japan in 1968, and the pathogen was initially identified as Phytophthora megasperma based on morphological characteristics. Similar Phytophthora isolates have since been collected from rose plants in Chiba, Kanagawa, and Shizuoka Prefectures. In 2005, several Phytophthora isolates were recovered from crowns of strawberry plants in Hokkaido Prefecture. These were considered to be members of a new species. In this study, we re-examined all these isolates using morphological and physiological studies and a multilocus phylogenetic analysis. The rose and strawberry isolates were mostly similar morphologically and physiologically, with some exceptions. The rose isolates differed significantly from P. megasperma sensu stricto and other related Phytophthora species. The rose and strawberry isolates had external proliferation of sporangia, characteristic funnel-shaped oogonia, predominantly paragynous antheridia, and fast growth rates of 10.5 mm/24 h at an optimum temperature of 28 °C. In the multilocus phylogenetic tree constructed using sequences from the rDNA ITS regions, rDNA LSU, and the translation elongation factor 1-α, β-tubulin and coxI genes, they formed a distinct monophyletic group in clade 7 with strong bootstrap support. The rose and strawberry isolates separated into two distinct groups. The results indicate that the rose and strawberry isolates constitute two separate species, designated here as Phytophthora nagaii and P. fragariaefolia.  相似文献

 在湖南省常德市西洞庭管区种植的朝鲜蓟(Cynara scolumus  L.)上发现了一种新的病害,其症状表现为地上部分从外层叶片开始逐步枯萎,随后根部和主茎杆的髓部腐烂变褐,最后整株枯萎。从田间感病朝鲜蓟茎杆的病健交界处用NA培养基分离,获得10个菌株,分别指定为HNXDT001~010,并进行了致病性测定、形态观察和细菌学特征分析,同时对HNXDT002菌株进行分子生物学鉴定。结果表明,该系列菌株在NA培养基上均形成灰白色圆形菌落,稍突起,有光泽,半透明。在显微镜下菌体呈短杆状,两端钝圆,具有2~8根周生鞭毛,革兰氏阴性。10个菌株通过针刺法接种均可导致朝鲜蓟茎杆、胡萝卜、辣椒、白菜、土豆、番茄和莴苣茎杆软腐,经科赫法则验证为致病病原菌。该菌株的16S rDNA序列和果胶酶基因片段测序(分别用16S rDNA通用引物16SF/16SR和果胶酶基因引物Y1/Y2扩增)与系统发育学分析表明,其16S rDNA序列(GenBank Accession No. JF721958)与胡萝卜软腐果胶杆菌胡萝卜亚种(Pectobacterium carotovorum  subsp.  carotovorum)菌株ATCC15713 (GenBank Accession No. U80197)序列同源性高达99%;果胶酶基因序列(GenBank Accession No. JF721960)与胡萝卜软腐果胶杆菌胡萝卜亚种(Pectobacterium carotovorum subsp. carotovorum)PC1菌株(GenBank Accession No. CP001657)序列同源性为93%。结果表明:朝鲜蓟细菌性根茎腐烂病病原为胡萝卜软腐果胶杆菌胡萝卜亚种。  相似文献

从海南5个芒果主产区采集分离芒果蒂腐病菌可可球二孢菌株,采用室内人工接种的方法对110个可可球二孢菌株进行致病力测定,采用区分剂量法和菌丝生长速率法测定其中106个菌株对多菌灵的抗性。结果表明:芒果蒂腐病自然发病率29.37%;人工接种的发病率100.00%,110株可可球二孢菌株中强致病力、中等致病力及弱致病力菌株分别占64.55%、25.45%和10.00%;在106株可可球二孢菌株中,抗性菌株发生频率为52.83%,其中高抗菌株可在含100 mg/L多菌灵培养基上生长,发生频率为45.28%,中抗菌株在含100 mg/L多菌灵培养基上不能正常生长,抗性频率为7.55%;没有发现低抗菌株。多菌灵对可可球二孢田间分离菌株EC50的频率分布为不连续分布,研究结果表明海南地区危害芒果的可可球二孢对多菌灵抗性非常严重。  相似文献

甘肃省马铃薯镰刀菌干腐病优势病原的分离鉴定   总被引:11,自引:0,他引:11  
 为明确甘肃马铃薯镰刀菌干腐病的优势病原,2006 年12 月~ 2007 年3 月由西至东从甘肃张掖、天祝、永登、临洮、渭源和西和等6 县市的马铃薯贮藏窖中采集表现有镰刀菌干腐病症状的马铃薯薯块,以组织分离法分离病原,单孢纯化镰刀菌(Fusarium spp. )菌株后,以形态学为基础,参照Nelson 镰刀菌分类系统进行鉴定。结果表明:6 个采样区共分离到293株镰刀菌菌株,其中以接骨木镰刀菌(F. sambucinum)和茄病镰刀菌(F. solani)出现频率高,是优势种。分析发现第一优势种随采样区而异,张掖、天祝和渭源采集的样品中茄病镰刀菌分离频率分别为42. 6% 、42. 1% 和32. 4% ,接骨木镰刀菌分离频率分别为14. 8% 、5. 3% 和26. 5% ,茄病镰刀菌为第一优势种;永登、临洮和西和采集的样品中接骨木镰刀菌分离频率分别为52郾1% 、50. 9% 和55. 2% ,茄病镰刀菌的分离频率分别为23. 3% 、32. 7% 和20. 7% ,接骨木镰刀菌为第一优势种。本文进一步对其在PDA、CLA 上的培养特征进行了观察和描述。按照柯赫氏法则用混合菌株接种法对大西洋(Atlantic)、夏波蒂(Shepody)以及一地方品种进行致病性测定,证实了优势菌种对块茎的致病性。利用EF鄄1琢基因引物(EF鄄1H 和EF鄄2T)对接骨木镰刀菌菌株GAUF鄄F12 进行基因组DNA 的PCR 扩增,将PCR 产物回收测序后在GenBank 上比对,菌株GAUF鄄F12 与GenBank 登记的接骨木镰刀菌5 个菌株的同源性均达99% ;用DNASTAR 分析软件将同源性较高的登记菌株的序列与GAUF鄄F12 菌株构建同源性树,结果表明:该菌株与以上5 个接骨木镰刀菌菌株均位于同源性树的同一分支,聚为一类,与形态学的鉴定结果一致。  相似文献
