The effect of fluctuation in water temperature during the freshwater rearing stage on cataract development in Atlantic salmon was investigated. Presmolts were exposed to either constant low or high temperatures, or high temperature with cold-water fluctuations. Temperature fluctuations caused the fastest growth rate and most severe cataracts, whilst low temperature resulted in slow growth and minor cataracts. The fish were then individually marked and maintained together in a seawater net pen for 7 months. Cataract progressed in all groups, fish from the low temperature group showed the fastest subsequent growth, and developed cataracts to the same degree as fish from the other groups. There were no statistically significant differences in selenium-dependent glutathione peroxidase levels in pooled samples of aqueous humour, lens and plasma from fish with or without cataract, except from the last freshwater samples, where cataractous fish showed lower plasma activity. Plasma cortisol and glucose levels did not differ, nor did whole eye riboflavin levels. Higher concentrations of some free amino acids in whole eyes were found in fish with cataract. This study shows that fluctuation in water temperature may cause both increased growth rate and cataract development in Atlantic salmon and that cataract development initiated in the freshwater rearing phase continues after transfers to seawater.  相似文献

The development of cataracts in Atlantic salmon, Salmo salar L., was studied in 16 groups of smolts fed diets differing in prooxidant (iron, copper, manganese) and antioxidant (vitamin E, vitamin C, astaxanthin) composition and lipid level for 23 weeks in sea water, using a 2(7-3) reduced factorial design. The seven dietary variables were systematically varied at low (requirement level and 150 g lipid kg(-1)) and high levels (below known toxic levels and 320 g lipid kg(-1)). A mean endpoint cataract incidence of approximately 36% was observed. High dietary levels of vitamin C and astaxanthin reduced cataract frequency, whereas high dietary lipid level, iron and manganese were associated with increased cataract frequencies. Considering the nutritional status of selected organs of the fish, only the status of ascorbic acid correlated negatively to cataract development (P < 0.05). The lens glutathione (GSH) status was not correlated to cataract frequency, nor statistically explained by the dietary variables. However, the study shows that balancing the diet with respect to pro- and antioxidant nutrients may significantly protect Atlantic salmon against development of cataracts. An incidence of reversible osmotic cataract observed at week 14 was positively correlated to plasma glucose concentration.  相似文献

The aim of the present study was to investigate cataract development in diploid (2N) and triploid (3N) Atlantic salmon smolts and post‐smolts at two water temperatures (10 and 16 °C) given diets with different histidine supplementation (LH, 10.4 and HH, 13.1 g kg?1) before and after seawater transfer. In freshwater, a severe cataract outbreak was recorded in both ploidies reared at 16 °C. The cataract score was significantly higher in triploids compared to diploids, and the severity was lower in both ploidies fed the HH diet. The cataract development at 10 °C was minor. Low gill Na+, K+‐ATPase activity in fish reared at 16 °C before seawater transfer was followed by osmoregulatory stress with elevated plasma electrolyte concentrations and high mortality in sea water. Both diploids and triploids reared at 10 °C developed cataracts during the seawater period, with higher severities in triploids than diploids and a reduced severity in the fish fed the HH diet. The findings of this study demonstrate the importance of environmental conditions in the husbandry of Atlantic salmon, and particularly triploids, with regard to smoltification and adjusted diets to mitigate cataract development in fresh and sea water.  相似文献

Abstract. Juvenile Atlantic salmon, Salmo salar L. were sampled from a commercial Norwegian fish farm during an outbreak of 'Hitra-disease'. One group of fish subjectively judged as healthy and another as diseased were defined on the basis of the classical apathetic behaviour seen in'Hitra-disease ď salmon. Haematological and biochemical analyses were carried out from blood and organs in 10 fish from each group. The diseased fish were severely anaemic. The blood indices MVC, MCH and MCHC indicated active erythropoiesis to compensate for the loss. Alkaline phosphatase (AP) activity, total protein, albumin, creatinine, triglycerides and total cholesterol were significantly reduced in the serum of diseased fish, while the activities of aspartate aminotransferase (ASAT) and alanine aminotransferase (ALAT) showed normal and significantly increased values respectively. Liver and spleen weight relative to the body weight and the content of water and lipid in the liver were significantly elevated in diseased fish. Furthermore, the iron content of the spleen was increased, whereas the zinc content showed no changes. The branched-chain amino acids valine, leucine and isoleucine were significantly higher and serine significantly lower in muscle extracts of diseased fish.  相似文献

Abstract. An outbreak of clinical furunculosis in commercially reared Atlantic salmon, Salmo salar L., broodfish in fresh water was successfully treated with a single intraperitoneal injection of oxytetracycline. The mean serum oxytetracycline residue in these fully mature fish did not decline and, at 8 weeks post-injection, was 3·7 μg/ml. In contrast, the mean serum residues in other fully mature but healthy fish declined during an 8-week experimental period to 1·3 μg/ml. The mean concentration of oxytetracycline in the liver of healthy mature fish was approximately two-fold that of similar aged but non-mature fish, and approximately four-fold the concentration found in post-smolts. Oxytetracycline residues 8 weeks after injection exceeded the minimum inhibitory concentration required for oxytetracycline sensitive Aeromonas salmonicida isolates in all groups, therefore under these circumstances, this antibiotic may be effective at a lower dose than that currently recommended. The histology of the liver from the furunculosis-surviving broodfish showed an increase in tissue lesions when compared with the other mature and non-mature fish in fresh water. The combination of prior infection and the physiological status of fasting broodfish may have influenced the rate at which this antibiotic is removed, and, in cases where these residues persist, there may be further damage to the liver.  相似文献

Effect of the eye fluke Diplostomum spathaceum on fish growth has remained somewhat unclear because 1) the question has not been subjected to experimental examination with treatment-control setup and 2) growth has not been related to the coverage of parasite-induced cataracts in a quantitative manner. We examined effects of the parasite on growth and competitive ability of whitefish (Coregonus lavaretus) in experimental conditions resembling those at fish farms by maintaining groups of exposed and control fish, as well as mixed groups from both treatments, under optimal conditions for 8 weeks. Contrary to our expectations, we did not observe differences in fish growth between the treatments. However, at the level of individual fish, parasite-induced cataracts had a negative effect on weight of the exposed fish but this took place only when the cataract coverage reached 100% in both eyes. No effect of the infection on competitive ability of fish was observed in mixed treatments which may be related to patterns of food distribution in the tanks or cues received by the fish from less infected conspecifics. Overall, the results suggest that effects of D. spathaceum on fish growth become apparent only in cases of very high cataract coverage, and that feeding regimes and the shape of the cataract coverage distribution in a fish group are important determinants of the magnitude of these effects.  相似文献

Diets, containing dehulled, solvent-extracted soybean meal in amounts corresponding to 0. 20 and 40% soybean protein of total protein, were fed for 10 months to duplicate groups of Atlantic salmon, Salmo salar L., weighing about 900 g, held in sea water. Fish fed the diet containing 20% soybean protein grew as well as fish fed the diet with high-quality fish meal as the sole protein source, whilst the fish fed the diet containing 40% soybean protein grew significantly less. No significant differences were found for dressing percentage, condition factor or relative liver weight between the dietary treatments. Fish fed the control or the 20% soybean meal (SBM) diets were not significantly different in body fat content, whilst this was significantly lower in the fish fed the 40% SBM diet. There were no significant differences between the dietary treatments in body protein content. Inclusion of SBM seemed to affect neither carcass calcium, carcass zinc, nor plasma calcium, magnesium or potassium. The plasma zinc content increased and plasma iron decreased by inclusion of SBM in the diets, probably reflecting dietary contents of zinc and iron. Liver iron was not affected. Plasma phosphate was significantly decreased by inclusion of SBM in the diet, whilst plasma glucose, total protein and haemoglobin were not affected. For plasma free amino acids, the only significant differences were a decrease in taurine with increasing SBM inclusion and methionine was significantly higher for the SBM groups than for the control groups.  相似文献

Abstract Infectious salmon anaemia (ISA) virus (ISAV) has been causing disease in New Brunswick since 1996. As a control measure, all fish in an outbreak cage are killed. The objective of this study was to compare ISAV prevalence in cages experiencing an outbreak with healthy cages from the same farm, neighbouring farms and distant farms. Atlantic salmon from five different groups were tested using an RT-PCR test. Groups included moribund fish from a cage experiencing an outbreak (A), healthy fish from an outbreak cage (B), healthy fish from a negative cage from a farm experiencing an outbreak in a different cage (C), healthy fish from a negative farm near an outbreak farm (D), and healthy fish sampled at a negative farm located in an area with only negative farms (E). Apparent prevalences (standard error) for the different groups (A-E) were 0.94 (+/-0.026), 0.41 (+/-0.062), 0.29 (+/-0.040), 0.08 (+/-0.037) and 0.08 (+/-0.037), respectively. All groups were significantly different (P < 0.002) from each other except for groups B and C and groups D and E. Because the prevalence of the virus was significantly higher in the outbreak cage (B) compared with other sites, early harvest of outbreak cages will remove one source of virus. However, ISA negative cages (C) that remain on the positive farm may potentially act as a viral reservoir.  相似文献

Abstract. This report describes the gross and histopathological appearance as well as the bacteriological findings in an outbreak of panophthalmitis in farmed chinook salmon. In all cases, the bacteria were either Mycobacterium neoaurum or an unidentified Rhodococcus spp.; in some fish, both types were present in roughly equal numbers, while in others, one type dominated. Despite occasionally recovering the bacteria from kidney or spleen, no lesions were found in these or in any other organ, suggesting that the localization to the eye was not incidental, but a specific tropism. Nevertheless, the possibility of predisposing cause(s) must be considered and these are discussed.  相似文献

Different chemical forms of essential minerals and trace elements supplemented to feed may show different bioavailability in fish. The present experiment was performed to compare an organic zinc form (Zn-gluconate) with zinc sulphate as dietary zinc sources for Atlantic salmon. Triplicate groups of fish were fed commercial fish meal based diets supplemented with either a low zinc level (50 mg Zn kg−1) or medium zinc level (180 mg Zn kg−1) as zinc gluconate or zinc sulphate for 6 months. Growth, mortality and feed conversion were recorded through the experiment. Fish were sampled initially and every month for zinc status analyses measured as whole-body zinc concentration. After 4 and 6 months, vertebral zinc concentration was measured and serum zinc was analysed at the end of the experiment. There were no differences in growth or mortality during the experiment. The results showed a significant effect of dietary zinc supplementation level on zinc status in fish for both zinc forms. However, there were no differences in zinc status between groups given different zinc forms. In conclusion, the present experiment showed that zinc sulphate and zinc gluconate are equally efficient as zinc supplements in salmon feeds within the concentrations normally found in salmon feed.  相似文献

A two-factor study was conducted to evaluate the effects of dietary riboflavin and lipid levels on the growth, health performance and riboflavin status of Atlantic salmon ( Salmo salar ). Atlantic salmon parr were fed four fishmeal-based diets with or without supplementation of 20 mg riboflavin kg–1, at two lipid levels, 150 or 300 g kg–1. Each diet was fed to triplicate tanks of fish for 12 weeks. Unsupplemented diets contained between 6 and 8 mg riboflavin kg–1. There were no significant differences in growth as a result of riboflavin supplementation. No mortality or histomorphological changes in eye tissues were observed. Dietary treatments did not affect blood haemoglobin values. After 12 weeks, muscle lipid content seemed to be reduced by riboflavin supplementation irrespective of dietary lipid level. Riboflavin status of whole body, muscle, liver, kidney and eye lenses is reported. Saturation levels of riboflavin in liver and muscle were reached with unsupplemented diets. The concentrations of riboflavin and lipid in liver were negatively correlated. There was a tendency of higher whole body riboflavin concentration in fish fed high-lipid diets. Based on growth, absence of deficiency signs and maximal tissue saturation of riboflavin, it can be concluded that the requirement for riboflavin was met by the natural riboflavin content in the raw materials of the feed. However, independent of dietary lipid level, dietary riboflavin supplementation may increase lipid utilization in rapidly growing salmon parr.  相似文献

We have previously documented increased survival by feeding tetradecylthioacetic acid (TTA) during a natural outbreak of infectious pancreatic necrosis in post-smolt S1 Atlantic salmon. The aim of the present study was to test the effects of dietary TTA in S0 smolt at a location where fish often experience natural outbreaks of heart and skeletal muscle inflammation (HSMI) during their first spring at sea. The experimental groups were fed a diet supplemented with 0.25% TTA for a 6-week period prior to a natural outbreak of HSMI in May 2007. Relative percent survival for the groups fed TTA was 45% compared with control diets, reducing mortality from 4.7% to 2.5%. Expression of genes related to lipid oxidation was higher in cardiac ventricles from salmon fed TTA compared with controls. In addition, salmon fed TTA had periodically reduced levels of plasma urea, and increased cardiosomatic index and growth. Reduced mortality and increased growth after administration of TTA may be related to a combination of anti-inflammatory effects, and an altered metabolic balance with better protein conservation because of increased lipid degradation.  相似文献

Growth was found to be associated with the changes of trypsin activity in the pyloric caecal tissues and the level of plasma insulin in Atlantic salmon (Salmo salar L.). A decrease in trypsin activity accompanied by an increase in plasma insulin was detected one month before an enhanced growth was observed. There were significant relationships between weight specific consumption rate, plasma insulin levels and fish growth. The correlation of weight specific consumption rate was higher with growth rate (R2=0.7, p<0.0001) than with plasma insulin concentration (R2=0.4, p<0.0001).When the comparison was made between Atlantic salmon carrying and lacking the trypsin variant TRP-2*92, the fish with the variant had lower maintenance ration (p<0.05), higher capacity for protein synthesis in the white muscle (p<0.02), and a greater ability to utilize the feed at a restricted ration than the fish without the variant. In Atlantic salmon lacking the variant, both plasma insulin concentrations and growth rates were significantly lower (p<0.05) in the fish fed 0.5% bw day–1 than those fed 1% bw day–1. Whilst the growth rates of TRP-2*92 salmon fed the different rations became similar one month after similar levels of plasma insulin were observed between them. The TRP-2*92 salmon may be defined as a high protein growth efficiency fish with low protein turnover rate.Genetic variation in trypsin isozyme pattern affects feed utilization, plasma insulin levels and growth in Atlantic salmon.  相似文献

Two size groups (0.5 and 3 kg) of Atlantic salmon, Salmo salar L., were stressed by netting and transport from sea cages into land-based tanks. Handling lasted for 1 h. Before onset of stress, basal concentrations of haematocrit and haemoglobin, plasma cortisol and glucose, and liver and muscle glycogen were measured. All levels were within ranges reported for salmon. After handling and at chosen time intervals during recovery, samples were taken to judge the impact of handling and whether secondary changes in carbohydrate metabolism were related of fish size. A low cortisol peak indicated a mild stress reaction after handling independent to fish size. Plasma glucose peaked as cortisol declined, and returned to basal levels within 48 h. Liver glycogen seemed to be the main source of plasma glucose. No changes were measured in muscle glycogen concentrations. The results indicate a high tolerance to handling stress in Atlantic salmon independent of fish size.  相似文献

In the last 9 years, epizootics of an icterus condition has affected coho salmon, Oncorhynchus kisutch (Walbaum), reared in seawater cages in southern regions of Chile. At necropsy, fish from field cases exhibited signs of jaundice accompanied by pale light-brown livers and dark spleens. Histopathological and haematological results indicated that these fish presented haemolytic anaemia. After microbiological examination no bacterial or viral agents could be identified as aetiological agents of this disease. In an infectivity trial, coho salmon, Atlantic salmon, Salmo salar L., and rainbow trout, Oncorhynchus mykiss (Walbaum), were inoculated intraperitoneally with a filtrate of an organ homogenate (0.45 microm) from a diseased coho salmon and held for 60 days in tanks supplied with fresh water. The disease was only reproduced in coho salmon in which mortalities, beginning at day 23 post-inoculation (p.i.), reached a cumulative value of 24% at day 27 p.i. This condition was transmitted to non-inoculated cohabiting coho salmon suggesting that it is a waterborne disease. Thus, this icteric condition is caused by an infectious form of haemolytic anaemia, probably of viral aetiology, and coho salmon are more susceptible than either Atlantic salmon or rainbow trout.  相似文献

Three duplicate groups of individually tagged lumpfish (mean initial weight: 20.0 ± 4.0 g) were fed three commercially available diets (A, B and C) for a period of 147 days. There were significant differences in growth rates between the groups with fish fed diet A having the highest growth rates whilst fish fed diet C achieved the lowest growth. Lumpfish fed diets A and B had lower prevalence and severity of cataracts compared with fish fed diet C, suggesting a possible dietary effect on cataract, alone or in combination with other environmental factors. There was a significant difference in the degree of liver vacuolization between the three dietary treatments at the end of the study period with fish fed diets A and B showing moderate to severe vacuolization compared with liver tissue from fish fed diet C, which had milder vacuolizations. There was little or no inflammation in pyloric caeca, midgut and hindgut and similarly for epithelial necrosis or vacuolization for all three dietary treatments at the end of the study. Fish fed diet A had the longest intestinal folds whilst fish fed diet C had the shortest.  相似文献

To study how hepatic lipid turnover and lipid transport may be affected by complete replacement of dietary fish oil (FO) with a vegetable oil blend (VO) from start feeding until the adult stages, Atlantic salmon (Salmo salar L.) were fed either 100% FO‐ or 100% VO‐based diets (55% rapeseed oil, 30% palm oil and 15% linseed oil) from start feeding until 22 months. Liver and plasma lipoprotein lipid class levels and lipoprotein fatty acid composition were analysed through the seawater phase, whereas liver fatty acid composition, plasma cholesterol, triacylglycerol (TAG) and protein levels were analysed through both freshwater and seawater stages. Further, enzyme activity of liver fatty acid synthetase (FAS), NADH‐isocitrate dehydrogenase, malic enzyme, glucose‐6‐phosphate dehydrogenase and 6‐phosphogluconate dehydrogenase and expression of the gene Peroxisome proliferator‐activated receptor γ (PPARγ) was analysed during both fresh water and seawater stages through the experiment. Dietary VO significantly increased salmon liver TAG and hence total liver lipid stores after 14 and 22 months of feeding. Further, after 22 months of feeding, plasma lipid levels and plasma low‐density lipoprotein (LDL) levels were significantly decreased in VO‐fed salmon compared with FO‐fed fish. The same trend, although not statistically significant, was seen for plasma very low‐density lipoprotein (VLDL). The activity of FAS was generally low throughout the experiment with the VO group having significantly lower activity after 16 months of feeding. The expression of PPARγ in livers increased prior to seawater transfer followed by a decrease, and then another increase towards the final sampling (22 months). Dietary vegetable oil replacement had no impact on PPARγ expression in salmon liver. In summary, liver TAG stores, plasma lipid and LDL levels were affected by dietary vegetable oil replacement in Atlantic salmon during a long–term feeding experiment. Current results indicate that high dietary vegetable oil inclusion increase hepatic TAG stores and decrease plasma lipid levels possible through decreased VLDL synthesis.  相似文献

The present study evaluated how different photoperiods, carbohydrate sources and inclusion levels influenced growth, plasma, organ and body compositions in Atlantic salmon (Salmo salar). Four different diets were used, containing 10 or 20% starch on a dry matter basis, using gelatinized corn (C10 and C20) or wheat (W10 and W20). Two different photoperiods were used, Continuous (Con) light throughout the study, Winter (Wi) two initial weeks of continuous light, thereafter 6 weeks of short day, ending with 6 weeks of continuous light. The winter regime resulted in reduced growth rate compared with continuous light. All Wi groups were smaller than Con groups when the experiment was terminated, except for fish given diet C10. Diet W10 resulted in better growth compared with all other groups when exposed to Con. regime. Atlantic salmon fed with diets containing 20% starch had lower growth rates than fish fed diets containing 10% starch, when the same starch source and light regime were compared. All fish belonging to the Con group exhibited similar and higher condition factors compared with fish from groups Wi. Condition factor was not influenced by dietary starch source. Higher whole body lipid concentrations were found when fish were given the corn compared with the wheat diets, without any responses to starch level or light regime. Whole body protein showed increased values in fish exposed to regime Wi compared with Con, without any influence from dietary manipulation. Liver glycogen values and hepatosomatic index reflected dietary starch levels and were significantly influenced by light regime, whilst muscle glycogen levels varied only as a response to light regime with lower values in fish from the Wi compared with the Con groups. Light regime influenced plasma glucose levels, except in groups fed with diet W10. All measured plasma nutrient concentrations were within normal ranges. At the end of the feeding period, all fish were evaluated for glucose and salt water tolerance to ascertain whether diet and/or light regime influenced glucose regulation capacity and/or smolt quality. Fish fed with 20% starch had reduced osmoregulatory ability measured as plasma chloride after a seawater challenge test. Fish from the Wi regime, but not from the Con regime developed characteristics associated with smoltification. Glucose tolerance in Atlantic salmon was substantially influenced by both photoperiod regime and diet. Fish reared under the Wi photoperiod showed reduced glucose regulation capacity compared with fish held under continuous light. Glucose regulation capacity was also reduced in fish adapted to the highest starch levels.  相似文献

If osmotic stress and reduced seawater tolerance are predisposing factors for infectious pancreatic necrosis (IPN) outbreaks in farmed Atlantic salmon, increased survival by enhancing access to energy would be expected. The aim of the present study was, therefore, to increase energy access in 1-year old Atlantic salmon after sea transfer by increasing the level of dietary fat, by exchanging some of the dietary oil with more easily oxidized medium chain triacylglycerols, or by dietary supplementation of potentially energy enhancing additives such as clofibrate and tetradecylthioacetic acid (TTA). A natural outbreak of IPN occurred 8 weeks after sea transfer, and a significant dietary effect explaining 76% of the variation in mortality was observed. Relative percentage survival for the fish fed TTA in sea water was 70% when compared with the unsupplemented control, reducing mortality from 7.8 to 2.3%. Muscle fat content and plasma chloride were related to IPN mortality, suggesting that reduced hypoosmoregulatory capacity might be a predisposing factor to the onset of an IPN outbreak. Based on the observation of a threefold increase in white muscle mitochondrial fatty acid oxidizing activity by TTA, it is suggested that TTA has resulted in a re-allocation of dietary fatty acids from storage to energy producing oxidation.  相似文献
