Pseudothecia of Pleospora allii developed best on garlic leaf debris infected by Stemphylium vesicarium incubated at low temperature (5–10°C) and relative humidity (RH) close to saturation. RH of less than 96% prevented the formation of pseudothecia, while an incubation temperature of 15–20°C led to the early degeneration of pseudothecia. Under natural conditions, colonization by pseudothecia of unburied garlic leaf debris varied between seasons from 6.0 to 15.5 pseudothecia/mm2, whereas lower colonization levels were recorded when samples were buried. Pseudothecial maturity was reached 1–4mo after the deposition of garlic debris on the soil surface and 15 days after the burial of residues. In the later case, pseudothecia degenerated with degradation of the plant debris. Ascospore release, which required rainfall or dew periods, occurred between late January and late April depending upon the year. A high correlation was found between pseudothecia maturation and four meteorological variables. Two of which, i.e. the number of hours with RH98% and with a mean temperature of 4.5–10.5°C, and the accumulated rainfall, explained most variability (adjusted R2=0.82–0.98 depending upon the year). A multiple regression equation relating the pseudothecia maturity index with these two variables could be used to forecast the epidemic onset of Stemphylium leaf spots in Southern Spain. Temporal progress of pseudothecia maturation was best fitted by a monomolecular model.  相似文献

The causal agent of a fohar disease of garlic ( Allium sutivum ) was identified as Stemphylium resicarium (Wallr) Simm Disease symptoms were reproduced on garlic leases 8 to 10 days after inoculation in a greenhouse (temperature range 22-26 C) Onion and tomato, two previously reported hosts of S, testcarnon. were also artificially infected with the isolate from garlic. Infection of onion by Alternaria porri and Stemphylium vesicarium was investigated under a range of controlled temperatures (4–25°C) and leaf wetness periods (0–24 h). Conidia of A. porri and S. vesicarium germinated within 2 h when incubated at 4°C. Terminal and intercalary appressoria were produced at similar frequencies at or above 10°C. The maximum number of appressoria was produced after 24 h at 25°C. Penetration of leaves by both pathogens was via the epidermis and stomata, but the frequency of stomatal penetration exceeded that of epidermal penetration. There was a strong correlation ( R 2 > 90%) between appressorium formation and total penetrations at all temperatures. Infection of onion leaves occurred after 16 h of leaf wetness at 15°C and 8 h of leaf wetness at 10–25°C, and infection increased with increasing leaf wetness duration to 24 h at all temperatures. Interruption of a single or double leaf wetness period by a dry period of 4–24 h had little effect on lesion numbers. Conidia of A. porri and S. vesicarium separately or in mixtures caused similar numbers of lesions. Alternaria porri and S. vesicarium are both potentially important pathogens in winter-grown Allium crops and purple leaf blotch symptoms were considered to be a complex caused by both pathogens.  相似文献

The incidence of garlic rot has constantly increased in France since the early 2000s. To set up an efficient method of garlic protection against this disease, we have clarified its aetiology. This was achieved by surveying garlic from the two main French basins of garlic production over 3 years. Fungi were isolated from 5,493 garlic cloves belonging to pink, purple, and white garlic types. Sequencing of the translation elongation factor 1α gene of 1,171 strains revealed that 94% of the strains belonged to the species Fusarium proliferatum and 6% belonged to F. oxysporum. The pathogenicity of both species on garlic was confirmed by artificial inoculations and reisolations. There was significantly more F. oxysporum in garlic cloves with symptoms coming from the southeast basin (9.44%) than from the southwest basin (2.76%). This study confirms that garlic rot is present in pink, purple, and white types. However, pink type garlic harbours F. oxysporum significantly less frequently (1.59%) than white (9.39%) and purple (7.34%) types. Sequencing of rpb1, rpb2, ITS, and IGS regions of a subsample of strains revealed that there is little genetic diversity in the French population of F. proliferatum.  相似文献

A total of 147 samples of garlic ( Allium sativum ) bulbs affected by blue mould were obtained from a variety of agroclimatic districts between December 1999 and February 2000. Penicillium species were identified using both morphological and chemotaxonomic characteristics. Penicillium allii was the predominant species isolated (81·8%) in this survey and the only species proven to be pathogenic on garlic. Other species were isolated much less frequently: P. chrysogenum (13·7%), P. brevicompactum (2·8%), P. phoeniceum (0·9%), P. aurantiogriseum (0·6%) and P. flavigenum (0·2%). Colonies of P. allii could be classified into four morphotypes and their distribution seemed to be influenced by seed trade and agricultural practices. Penicillium allii isolates were grouped into three aggressiveness phenotypes (low, medium and high) based on their ability to cause disease during field trials on susceptible (Fuego INTA) and less susceptible (Castaño INTA) garlic cultivars. The number of surviving plants at 191 days after planting and postharvest bulb weight contributed the most towards aggressiveness modelling.  相似文献

Ascospores and conidia released into the air were recorded around plots on which garlic debris infected by Stemphylium vesicarium were fixed onto the soil surface. Symptoms in garlic trap plots located in the vicinity of infected debris, started in March and developed during April–May to reach disease incidence close to 100%, final disease severity values being lower in 1993 and 1995 than in 1994 and 1996. Whereas daily concentrations of ascospores were rather erratic, with 30% of captures between 0 and 6 h, conidia showed a daily periodicity with highest concentrations between 12 and 18 h, with a pronounced peak between 14 and 16 h, and lowest values at night. Ascospore release occurred mainly in February and March. It coincided with rainfall periods, 14 h with vapour pressure deficit 5 mb and solar radiation <145 W m–2 on the current day of the capture. In contrast, greatest captures of conidia started in late April and were prevalent in May, and were associated with rainfall in days previous to the capture in which rather high temperature occurred and solar radiation was 109–345 W m–2. Among the weather variables considered, rainfall appeared directly related to the aerial concentration of ascospores and conidia. The role of relative humidity seemed essential when rainfall did not occur. There was a relationship between conidia concentration in the air and number of hours with temperature in the range 12–21 °C. Ascospore production was not essential for infections to take place, since primary infection from conidia may occur and disease can develop from them readily.  相似文献

Crop loss of onion bulbs during storage carries an exceptionally high economic impact because a large portion of the production expenses has been expended before storage. Because of this, it is important to define practices that can reduce onion bulb losses caused by storage rots. This study investigates the impact of various curing parameters on disease development resulting from infection by Pantoea agglomerans, P. ananatis and P. allii on onion bulb cultivars Vaquero and Redwing, during storage. Overall, both the incidence and mean rot severity were similar amongst the bulbs under comparable conditions regardless of the species of Pantoea inoculated, although a significant difference was detected between the two onion bulb cultivars. In addition, a significant reduction of storage rot was observed when curing temperatures were ≤35°C. At temperatures >35°C, a shorter curing duration (2 days vs 14 days) decreased the severity of bulb rot due to Pantoea. This increased understanding of the inter‐relationships between the parameters used for curing, and the incidence and severity of bulb rot caused by Pantoea helps provide guidance towards using the curing process as a means to reduce the level of damage resulting from post‐harvest storage rot.  相似文献

 本文对来自红海榄(Rhizophora stylosa)赤斑病同一病斑上获得的2个菌株PⅠ-1和PⅡ-3进行了鉴定和生物学特性研究。结果表明,PⅡ-3菌株的致病力比PⅠ-1的稍强;2个菌株均具有一定的寄主专化性。经形态特征观察与rDNA-ITS序列分析,2个菌株均鉴定为异色拟盘多毛孢(Pestalotiopsis versicolor)。生物学特性测定结果表明,温度、pH值和光照对2个菌株菌丝生长及孢子萌发的影响差异不大,生长温度均为10~32℃(最适25℃),pH值为3.0~10.9(最适4.0);孢子萌发温度均为10~35℃(最适25~28℃),pH值为2.5~8.0(最适3.0);黑暗条件有利于2个菌株菌丝生长和孢子萌发。但2个菌株的产孢特性存在明显差异,其中PⅠ-1产孢的最适温度为28℃而PⅡ-3为20℃;PⅠ-1产孢的最适pH值为5.1而PⅡ-3为9.0;PⅠ-1产孢对光照不敏感,而PⅡ-3在黑暗条件下几乎不产孢。另外,在相似条件下,PⅠ-1产孢量比PⅡ-3大,而PⅡ-3菌丝生长速度比PⅠ-1快,孢子萌发率也比PⅠ-1高。由此可见,2个菌株PⅠ-1和PⅡ-3虽属于同一个种,但某些生物学特性存在一定的差异。  相似文献

为了明确河南省洛阳地区牡丹叶部一种新病害的病原种类,对采自田间的典型症状叶片进行了常规组织分离,对单孢菌株进行了形态鉴定、rDNA ITS序列分析及致病性测定.结果表明,病原菌在PDA培养基上菌落初期白色绒毛状,后期红褐色.分生孢子梗通常呈扫帚状,有2~3个分支,末端膨大呈泡囊状.分生孢子直棒状,两端钝圆,有0~1个隔膜,大小为(38.4~86.4)μm× (4.6~5.9)μm,平均60.1 μm×5.2 μm.离体叶片与活体植株接种结果表明,该菌均能侵染牡丹叶片并引起与田间相同的症状.据此将分离物鉴定为Cylindrocladium canadense,将该病害命名为牡丹柱枝孢叶斑病.  相似文献

通过症状观察、致病性测定、形态和培养性状观察、rDNA-ITS序列分析及生物学活性测定将分离到的病原菌鉴定为异色拟盘多毛孢Pestalotiopsis versicolor(Speg.)Steyaert。菌株JG01在PDA培养基上菌落白色、背面呈橘黄色花瓣状,分生孢子盘墨汁状散生在菌丝上,分生孢子长梭形,大小为18.3~27.4μm×5.6~7.7μm,4隔,中间3个细胞着色,分隔处颜色很深,顶端附属丝一般2~3根。ITS序列分析,菌株JG01与P.versicolor(AF405298)亲缘关系最近,同源性达到99.60%。在PDMA培养基上,25~28℃、pH 4.0~6.0、碳氮源分别为葡萄糖和酵母提取物的条件下菌丝生长最快,培养条件对菌株产孢的影响不明显。  相似文献

 本研究通过致病性测定、形态学和分子生物学鉴定方法,首次明确陕西省发生的玉米条斑型叶斑病是玉米圆斑病的一种症状类型,由玉米生离蠕孢(Bipolaris zeicola)3号生理小种引起。供试品种豫玉22、农大108、农大3138、东单7号、沈单10号、沈单16号、户单4号、新陕资1号、高农1号和陕单9号都对3号生理小种表现感病,品种间抗病性差异不显著。此菌在PDA、PSA、OMA、CMA、WA和Czapek上均可生长,最适生长培养基为PDA。菌落生长温度为5~35℃,最适菌落生长温度为25℃。分生孢子在5~40℃均可萌发,低于5℃高于40℃萌发率极低,萌发的最适温度为28℃。孢子在pH 3~11均可萌发,萌发最适pH为5。在自来水中分生孢子可以萌发,蔗糖和葡萄糖溶液可以明显促进孢子萌发。  相似文献

 为明确黄淮海夏玉米区玉米灰斑病的病原菌种类,于2017年从河北、河南、山东、安徽、天津五省市疑似发生玉米灰斑病的76个市县采集样本,采用组织分离法和显微单孢分离法获得病原菌,通过形态与分子生物学手段进行病原菌种类鉴定,并对黄淮海地区、陕西、辽宁、黑龙江的37个菌株进行ISSR聚类分析。结果表明,引起黄淮海地区玉米灰斑病的病原菌为玉蜀黍尾孢(Cercospora zeae-maydis),玉蜀黍尾孢的变异具有地理种群特征。同时对黄淮海夏玉米区主栽品种进行人工接种鉴定,结果显示6个主栽品种中郑单958、登海605、先玉335、浚单20等4个品种对玉米灰斑病抗性较差,玉米灰斑病在本区域具有大面积发生和流行的风险。本研究为黄淮海夏玉米区灰斑病的综合防控提供支撑和理论依据。  相似文献

对11份玉米常用骨干自交系及8份重点推广的玉米杂交组合的抗病性进行苗期人工接种鉴定。结果表明,试材之间抗性差异较大,抗性较好的试材有沈137、Mo17、799、沈试29、丹玉13和丹408等。杂交组合的抗病性与母本及父母本中亲值相关密切,相关系数r分别为093和094,而与父本相关系数r为017,母本的抗病性对杂交组合的抗病性决定程度较大。  相似文献

In organic seed production of Brassica vegetables, infections by Alternaria brassicicola and A. brassicae can cause severe losses of yield and seed quality. Four field experiments with or without artificial inoculation with A. brassicicola were conducted in organically managed seed‐production crops of cauliflower cv. Opaal RZ in 2005 and 2006 in the Netherlands. The development of A. brassicicola and A. brassicae on pod tissues and developing seeds was followed and seed quality was assessed. Alternaria brassicicola was externally present on 1·2% of the seeds 14 days after flowering and observed internally within 4 weeks after flowering. In both seasons, seed colonization by the pathogen increased slowly until maturation but sharply increased during maturation. A similar pattern was found for the colonization of pod tissues by A. brassicicola as quantified by TaqMan‐PCR. The incidence of A. brassicicola on mature seeds reached 70–90%. Internal colonization was found for 62–80% of the seeds. External and internal seed colonization by A. brassicae was much lower, with incidences below 3%. The quality of harvested seeds was generally low, with less than 80% of seeds able to germinate. Seed quality was not affected by warm water treatments. It was concluded that A. brassicicola and A. brassicae have the potential to infect pods and seeds soon after flowering. For the production of high quality seeds, producers must prevent such early infections. Therefore, new control measures are needed for use in organic cropping systems.  相似文献

应用离体叶片点滴接种平脐蠕孢菌Bipolaris sorokiniana孢子悬浮液并进行电镜观察,研究小麦及其近缘种属叶片上病斑型的发生特点及病、健组织细胞内部超微结构的变化。结果表明,不同品种小麦相同发育阶段的离体叶上病斑型的变化仅与其自身抗病性遗传表达有关。小麦病斑的反应型变化过程,随时间在品种、叶位、不同发育阶段间存在差异,叶肉病变细胞中细胞器结构及功能完好的为14.3%,变化的为28.6%,解体和功能丧失的为57.1%;小麦近缘种病斑的反应型不随时间改变,仅在种属间存在差异,且叶位间的差异不显著,大赖草病变细胞中细胞器结构及功能完好、改变和丧失的均为33.3%。  相似文献
