利用废革屑制备肽Ca螯合物的工艺探究   总被引:1,自引:0,他引:1  
本论文通过水解革屑制备多肽水解液,并将水解液与Ca Cl2进行螯合反应制备多肽-Ca金属螯合物。对水解液进行理化性能进行表征,利用GPC测定水解液的分子量;设计单因素螯合实验及正交试验,考察了p H、温度、时间、肽/Ca质量比等因素对螯合反应的影响,确定了最佳反应条件。通过研究发现不同因素对螯合率的影响大小为:温度时间肽Ca比 pH;对得率的影响大小为:pH温度肽Ca比时间。最佳螯合条件:反应时间90 min,肽Ca比为4∶1,反应温度为70℃,pH为7,最佳条件下的螯合率为86.55%;最后,对肽Ca螯合物进行傅里叶红外光谱、扫描电镜及能量色散X射线光谱仪分析表征,结果表明Ca离子成功螯合到多肽上,得到Ca肽螯合物。  相似文献

为了充分利用猪骨资源,该文以猪骨粉为原料,研究了胶原多肽螯合钙的制备工艺。首先利用乳酸菌发酵酶解猪骨粉,以发酵液中游离钙含量为指标,确定最佳发酵条件为:蔗糖添加量7%,温度35℃,转速120 r/min,接种量12%,发酵时间18 h。对发酵液进行浓缩后,再与胶原多肽溶液进行螯合,通过单因素和响应面试验,确定螯合的最优条件为:pH值7.9,温度40℃,胶原多肽溶液与发酵液体积比为7∶1,螯合时间 4 h,无水乙醇添加倍数为11,此时螯合率为79.9%,为猪骨的深入开发提供依据。  相似文献

采用45Ca示踪 ,45Ca显微放射自显影和X 射线能谱分析技术 ,探索苹果幼果组织钙运输途径及激素对该途径的调控方式。结果表明 ,表皮层并未完全阻碍Ca2+由质外体进入果肉组织 ,Ca2+可通过质外体和共质体两种途径在幼果组织中运输 ,以质外体途径为主 ;IAA、GA和NAA虽对幼果组织Ca2+的运输均有明显的促进作用 ,但机理不同。GA主要促进Ca2+由质外体途径进入幼果组织 ,而IAA和NAA主要促进幼果中Ca2+的共质体途径运输。  相似文献

以谷氨酸和硫酸锌为原料,制备谷氨酸锌螯合物。用有机溶剂沉淀法,以螯合率为指标,筛选最佳螯合及分离提纯条件。结果表明,最佳螯合条件为:pH 6.5,螯合温度90℃,反应时间30 min;分离最佳条件为:将甲醇与螯合浓缩液按10∶1的比例混合提取谷氨酸锌螯合物,用20%的硫酸溶解该螯合物。试验证明,该条件下谷氨酸螯合锌螯合率可达80%以上。此外,定性试验结果均表明谷氨酸与硫酸锌发生了螯合反应。说明该试验条件下可制得稳定且具较高螯合率的谷氨酸锌产品。  相似文献

为了优化蟹肉酶解肽的锌螯合工艺,以细点圆趾蟹肉为原料,制得蟹肉酶解物,通过单因素试验,考察酶解物与硫酸锌的质量比、pH、反应时间、酶解物浓度、反应温度对锌修饰蟹肉酶解肽的影响,采用正交试验优化蟹肉酶解肽的锌修饰工艺。结果表明,在酶解物与硫酸锌的质量比2∶1、pH值6.5、时间60 min、酶解物浓度5%、温度55℃条件下,酶解肽-锌的螯合率为91.52%,螯合物得率为25.83%,该工艺条件下酶解肽与锌的螯合率和螯合物得率均较高。本研究结果为新型生物态锌的研制、细点圆趾蟹的高值化利用提供了依据。  相似文献

以废弃鸡毛水解液为复合氨基酸来源,研究其与微量元素铁、锌、锰、铜螯合的最佳工艺条件,并利用有机溶剂沉淀分离法制备固体复合氨基酸亚铁、锌、锰、铜螯合物,以螯合率为指标考察影响制备工艺的主要因素。结果表明,最佳工艺参数为:复合氨基酸与亚铁、锌、锰和铜螯合反应的最佳配位体摩尔比均为2∶1;最佳pH值范围分别为5.0、8.0、4.0~5.0和6.0~7.0;反应温度为室温(222~6℃);反应时间为30.min。在上述条件下,复合氨基酸与各微量元素的螯合率均超过95%;所得复合氨基酸螯合物可与0.2.mol/L磷酸二氢钾复配,克服了传统微肥完全不能与磷酸盐复配的缺陷。  相似文献

为探究不同山梨醇螯合钙喷施浓度对花生生长及养分吸收的影响,本研究采用大田实验,以花育22号为试材,在施基肥基础上设置5个钙素喷施浓度处理 (以Ca2+计,单位g/L):0 (CK)、1.4 (T1)、1.6 (T2)、1.8 (T3)与2.0 (T4),测定了各处理对花生光合作用、收获期产量、品质、各器官干物质积累量及钾、钙、镁含量。结果表明,(1)与CK处理相比,喷施山梨醇螯合钙提高了叶片SPAD值,促进了花生光合作用,尤其在结荚期;(2)喷施山梨醇螯合钙能提高花生产量及粗脂肪、粗蛋白含量,其中T2处理较CK处理分别提高11.48%、5.74%、7.82%;(3)施钙能够显著增加籽仁干物质积累量,有效提高花生生殖器官干物质占比,降低营养器官干物质分配比例,以T3处理改善效果较为突出,其次是T2处理;(4)喷施山梨醇螯合钙对花生各器官钾、钙、镁含量的影响不同,但总体上促进了花生对养分的吸收。主成分分析结果表明在基础施肥前提下喷施山梨醇螯合钙显著影响了花生生长发育。综合来看,喷施钙离子浓度为1.6 ~ 1.8 g/L的山梨醇螯合钙对花生增产提质效果更为突出。  相似文献

研究不同浓度钙(0、1和10 mmo1/L CaCl2 ;5 mmo1/L EGTA)对苹果果实钙调蛋白(CaM)含量和Ca2+-ATPase活性及其基因表达的影响。利用同源克隆方法分离CaM和Ca2+-ATPase基因,采用荧光定量PCR方法研究它们表达特征。结果表明,果实切片外源补钙,可溶性Ca2+及CaM含量在高钙处理12 h达到高峰;高钙处理12 h质膜Ca2+-ATPase活性显著增加,与胞内CaM含量增加时间一致;高钙处理24 h液泡膜Ca2+-ATPase活性显著增加;随着质膜和液泡膜Ca2+-ATPase活性显著增加,可溶性Ca2+含量在加钙处理48 h显著下降。研究基因表达发现,加钙处理6 h苹果CaM基因的表达量显著增加;加钙处理12 h苹果Ca2+-ATPase基因的表达量显著增加,与CaM含量及质膜Ca2 +-ATPase活性变化一致。果实缺钙处理显著增加CaM基因表达量,而苹果Ca2 +-ATPase基因的表达量没有显著变化。上述研究表明,苹果补钙可以提高细胞内可溶性Ca2+和CaM含量以及CaM基因的表达量,有效启动钙信使系统;质膜及液泡膜Ca2+-ATPase是调控胞内Ca2+关键的酶,通过提高质膜及液泡膜Ca2+-ATPase的活性及Ca2+-ATPase基因的表达量,维持胞内Ca2+的稳态水平。  相似文献

采用45Ca微观放射自显影、电子探针及特异性抑制剂研究花生荚果钙素吸收机制,结果表明,Ca2+是通过主动吸收由外界进入细胞质中,并以共质体途径在组织和细胞间运输,外果皮的周皮层和中果皮的纤维细胞层对Ca2+质外体运输有一定阻碍作用。钙通道抑制剂与ATP酶特异性抑制剂处理,中果皮和内果皮的薄壁细胞内未见45Ca显影,大量45Ca出现在周皮层,仅微量的45Ca到达中果皮细胞间隙和纤维细胞层质外体空间。电子探针的结果也可看出高钙峰出现在周皮层,而对照处理荚果整个组织的共质体和质外体均有45a的显影,且由外果皮向内,组织中的钙呈逆浓度梯度分布。2,4二硝基酸可抑制荚果钙素吸收速率,其抑制率达7092%。荚果钙素吸收动力学的结果表明,当Ca2+浓度为0-0.5mmol/L时,其吸收速率符合Michaelis-Menten酶动力学模型,Km值为0.0135mmol/L,Fmax为13210-4.mol/(cm2h);而当Ca2+浓度为1-5mmol/L范围时,其表现出复杂的吸收特征,此时Km和Fmax均无法得出明确的数值;供钙浓度为0.5-2.0mmol/L时,荚果干重及果仁干重与吸钙量均可达到最大并趋于稳定。  相似文献

有机酸在岩石矿物溶蚀风化及土壤形成的过程中具有较强的促进作用,了解柠檬酸对珊瑚砂的溶蚀过程对我国岛礁建设具有重要意义。本文通过不同浓度、不同pH柠檬酸溶液对珊瑚砂的溶蚀实验,研究柠檬酸对珊瑚砂的溶蚀过程,探讨其溶蚀机理。结果表明,在不同浓度柠檬酸溶液中,反应初期珊瑚砂中的Ca2+、Mg2+迅速溶出,随后大量的Ca2+与柠檬酸反应生成柠檬酸钙沉淀。平衡时溶液中Ca2+、Mg2+浓度随着柠檬酸浓度的增加而增加。在不同初始pH柠檬酸溶液中,当pH<7时,溶液中Ca2+、Mg2+的浓度随着初始pH的升高而降低,Ca2+、Mg2+主要由碳酸盐矿物与H+反应而溶出。当初始pH≥7时,Ca2+、Mg2+与柠檬酸生成络合物促进Ca2+、Mg2+的溶出,随着pH的升高,Ca2+、Mg2+浓度略有降低。珊瑚砂的溶蚀与柠檬酸钙的沉淀结晶主要受pH控制。通过XRD和SEM的分析表明,高浓度柠檬酸与珊瑚砂反应生成片状的柠檬酸钙沉淀,且饱和溶液中柠檬酸与钙离子的结晶生成平整规则的条状四水合柠檬酸钙沉淀。珊瑚砂溶蚀过程中镁方解石最先发生溶蚀,其次为文石,最后为方解石。  相似文献

【目的】以富士(Fuji)、 秦冠(Qinguan)嫁接在平邑甜茶(Malus hupehensis Rehd.)上的当年生盆栽苗为试验材料,采用砂培方法,研究了缺氮胁迫和干旱对富士和秦冠生长情况、 光合参数、 植株各部位氮磷钾含量及氮素利用效率的影响,分析比较了低氮干旱条件下富士和秦冠生长及氮素利用的差异,以期为果树生产高效肥水利用提供理论指导。【方法】试验共设四个处理: 正常氮正常水(ZZ)、 低氮正常水(DZ)、 正常氮干旱(ZG)、 低氮干旱(DG)。氮素和水分均设置两个水平,分别为正常氮(6 mmol/L NO-3-N)、 低氮(0.3 mmol/LNO-3-N)、 正常供水(保持盆中砂子相对含水量为饱和含水量的80%~85%)、 干旱处理(保持盆中砂子相对含水量为饱和含水量的60%~65%)。【结果】富士和秦冠的生物量(茎和叶)、 株高茎粗等生长指标以及光合速率、 气孔导度、 蒸腾速率均为正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG),并且相对应处理下秦冠的以上指标均高于富士;正常供水下,缺氮处理使富士、 秦冠的根冠比比正常氮处理均有所增加,富士提高了2.05%,秦冠提高了22.40%。富士和秦冠的氮、 磷、 钾含量均表现出正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG); 氮、 钾元素含量在植株各部位的分布顺序依次是叶>根>茎,磷元素则是根>叶>茎;光合氮素利用效率(PNUE)和氮素利用效率表现为秦冠处理之间差异极显著,富士处理之间差异不显著;秦冠的PNUE和NUE明显高于富士,在低氮正常水(DZ)处理下,秦冠氮肥利用率比富士高42.07%,在低氮干旱(DG)处理下高64.14%;低氮胁迫下富士和秦冠的NUE显著提高,并且秦冠提高的幅度高于富士。【结论】施用氮肥能够显著提高富士与秦冠的干物质量,同等水肥条件下,秦冠生长优于富士;水分亏缺会减少叶片对氮的吸收,干旱条件下适度增施氮肥,可提高果树的抗旱能力;低氮干旱胁迫下秦冠的生长指标、 光合指标及氮素利用效率指标均优于富士,表现出较强的抗低氮干旱胁迫的能力。  相似文献

Staff members of the Department of Botany of Palacký University in Olomouc and Gene Bank Department – Workplace Olomouc, Research Institute of Crop Production in Prague, Czech Republic, conducted an expedition in seven European countries (Austria, Czech Republic, France, Germany, Italy, the Netherlands, Switzerland) in August/September 1999 to collect wild Lactuca spp. germplasm and study its geographic distribution, ecology and biodiversity. During the mission, more than 600 locations were visited resulting in the collection of 602 seed samples (accessions) of wild Lactuca species and 13 seed samples of related genera (Chondrilla and Mycelis). Lactuca serriola f. serriola, L. serriola f. integrifolia, L. saligna and L. viminea subsp. chondrilliflora were prevalent in southern Europe (Italy, France), however, only L. serriola was common in central and western Europe (Austria, Czech Republic, Germany, Netherlands, Switzerland). The greatest diversity of Lactuca species was found in France, where also the most seed samples (165) were collected. The most characteristic habitats with a high density of Lactuca spp. populations were observed along roads and highways, grassy ditches, ruderal communities, and dust-heaps. Natural infections by powdery mildew (Erysiphe cichoracearum) and downy mildew (Bremia lactucae) on some wild Lactuca spp. were observed. Recent observations concerning the geographic distribution, population structure, habitats, and natural occurrence of diseases of Lactuca spp. are discussed. This assemblage of genetic resources of Lactuca spp. can serve as the basis of future studies of species diversification, spatial population structure, plant microevolution, domestication processes, and genetic variability of host-parasite interactions.  相似文献

Laser-induced breakdown spectroscopy (LIBS) is a new technique for the analysis of plant material. This study investigates the application of LIBS to pasture-based plant samples. The LIBS measurements were obtained from pelletized pasture samples (100 samples) that had been also analyzed by inductively coupled plasma–optical emission spectroscopy (ICP-OES) following microwave digestion for calibration and comparison purposes. Comparisons for elements sodium (Na), potassium (K), magnesium (Mg), calcium (Ca), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn), boron (B), phosphorus (P), and sulfur (S) showed that LIBS could be used for almost all the standard profile total elements with concentrations down to low mg/kg levels (observed error of Na: 0.024 percent, K: 0.18 percent, Mg: 0.016 percent, Ca: 0.073 percent, P: 0.017 percent, Mn: 31 mg/kg, Fe: 150 mg/kg, Zn: 6.6 mg/kg, and B: 1.1 mg/kg). Elemental analysis at less than mg/kg levels was not possible using LIBS. The elements S and Cu were particularly difficult to analyze with reliability using LIBS at the concentration levels found in the plant samples. Replacing microwave digestion and subsequent ICP analysis with a direct analysis of dried plant samples using LIBS has the potential to improve the productivity and reduce the cost of testing.  相似文献

We have studied the effect of anaerobically digested (ADMSW) and composted municipal solid waste (CMSW) on mineralization and foodweb dynamics to verify the hypothesis that ADMSW would immobilize N right after addition to soil in contrast to addition with CMSW. Another hypothesis was that the mesofauna (enchytraeids and microarthropods) would stimulate N release from the decomposer community. We measured excretion of -N and urea-N from the mesofauna and hypothesized that enchytraeids would release urea. ADMSW and CMSW were amended to pots with sandy loam and barley. The pots were divided into treatments with or without mesofauna. Mesofauna, plant N and biomass, soil N and ergosterol (fungal biomass) were measured over a 113-day period of four equidistant samplings. Soil respiration, N mineralization and N release by the mesofauna were modelled from concurrent studies. ADMSW- and CMSW-treated soils initially (<20 days) immobilized N. The amendments did not increase plant growth substantially, and this was probably due to N-limitation in the early stages of plant growth. Enchytraeid abundance was about three times higher in ADMSW- than CMSW-treated soils, indicating that ADMSW contained more labile compounds, bacteria, and microfauna. The mesofauna did not affect N-content, but the cumulated -N excreted by the mesofauna was estimated to be substantial and about one-fifth of total plant N in ADMSW. An explanation for this discrepancy might be that in the absence of the mesofauna, other members of the detrivore and microbivore community performed the mesofauna’s function. Neither enchytraeids nor microarthropods excreted urea.  相似文献

The responses of tallgrass prairie plant communities and ecosystem processes to fire and grazing are well characterized. However, responses of invertebrate consumer groups, and particularly soil-dwelling organisms, to these disturbances are not well known. At Konza Prairie Biological Station, we sampled soil macroinvertebrates in 1994 and 1999 as part of a long-term experiment designed to examine the effects and interactions of annual fire, mowing, and fertilization (N and P) on prairie soil communities and processes. For nearly all taxa, in both years, responses were characterized by significant treatment interactions, but some general patterns were evident. Introduced European earthworms (Aporrectodea spp. and Octolasion spp.) were most abundant in plots where fire was excluded, and the proportion of the total earthworm community consisting of introduced earthworms was greater in unburned, unmowed, and fertilized plots. Nymphs of two Cicada genera were collected (Cicadetta spp. and Tibicen spp.). Cicadetta nymphs were more abundant in burned plots, but mowing reduced their abundance. Tibicen nymphs were collected almost exclusively from unburned plots. Treatment effects on herbivorous beetle larvae (Scarabaeidae, Elateridae, and Curculionidae) were variable, but nutrient additions (N or P) usually resulted in greater densities, whereas mowing usually resulted in lower densities. Our results suggest that departures from historical disturbance regimes (i.e. frequent fire and grazing) may render soils more susceptible to increased numbers of European earthworms, and that interactions between fire, aboveground biomass removal, and vegetation responses affect the structure and composition of invertebrate communities in tallgrass prairie soils.  相似文献

“Wicked” problems are those that are complex and that change when solutions are applied. Many conflicts in conservation fall into this category. The study approached the problem of how to constrain the apparent wickedness of a problem in the conservation management of a species by using simple empirical indicators to carry out iterative assessment of the risk to a population and to document how this risk evolves in relation to the addition of new data and the implementation of management actions. Effects of high levels of uncertainty within data and also concerning population structure were examined through stochastic simulation and by exploration of scenarios. Historical trends in the example used, the Steller sea lion, showed rapid declines in abundance in some regions during the 1980s. The current total population is 130,000-150,000 Steller sea lions through Alaska and British Columbia and this number has been stable since about 1990 in spite of regional differences in population dynamics. Regional differences in the sequence of changes in the number of pups and non-pups, suggested that an internal re-distribution of juveniles could have happened between 1980 and 1990. Current productivity also appears close to the long-term mean. Stochastic population projection using various scenarios showed that, based upon this history, the risk of extinction for the population has declined and is below reasonable thresholds for considering the population to be endangered. The trends in risk suggest that management actions taken since 1990 have probably been effective. Consequently, the conservation management objectives for the Steller sea lion are probably being met. The approach provides a mechanism, based upon experience and scenario analysis, for exploring future policy options and may help to constrain the debate amongst stakeholders about the cost-benefit trade-offs associated with different options.  相似文献

Nutrient distributions under no tillage (NT) compared with conventional disk-and-bed tillage (CT) management in the warm, humid region of the southeastern USA need to be assessed so that future placement, quantity, and type of fertilizers can be altered, if necessary, to efficiently match crop demands. We determined soil-profile distributions of pH, N, P, S, K, Ca, Mg, Na, Zn, Fe, Mn, and Cu to a depth of 0.9 m at the end of 8.5 years of continuous CT and NT management on a Weswood silty clay loam (fine, mixed, thermic Fluventic Ustochrept) in southcentral Texas. Most dramatic changes occurred within the 0–0.05 m depth, where soil under NT had lower pH, Fe, and Cu than under CT, but greater P, K, Zn, and Mn. Greater P and K under NT than under CT also occurred below the till-zone (0.15–0.3 m). At a depth of 0–0.3 m, soil under NT contained greater amounts of extractable P, K, Zn, Fe, Mn, and Cu than under CT. Nitrogen fertilization had little effect on nutrient distributions, except resulting in greater extractable K at 0–0.05 m and greater nitrate at 0–0.15 m. Few changes in soil-profile distributions were observed for extractable S, Ca, Mg, and Na. Long-term continuous use of NT on this fine-textured, high-fertility (except for N) soil had no apparent adverse effects on nutrient distributions relative to CT, but enhanced conservation and availability of P, K, Zn, Fe, Mn, and Cu near the soil surface where crop roots proliferate.  相似文献
