Although we recently clarified sex-based differences in mouse kidney morphology, the developmental processes responsible for these gender differences during maturation remain unclear. The present study analyzed the morphometry of kidneys from 20-, 30-, 50-, 60-, 70-, 90-, 120- and 150-day-old DBA/2Cr mice. Total kidney weight and ratio of kidney weight to body weight were larger in males than females beginning at 50 days of age. The percentage of renal corpuscles exhibiting a cuboidal parietal layer was higher in males than in females in the 70-day and older mice. The diameter of cortical renal corpuscles was larger in males than in females beginning on day 90. The number of proximal convoluted tubular cell nuclei was higher in females than in males from day 90 onward. Vacuolar structures in the proximal convoluted tubular epithelium became prominent in 70-day-old males. PAS-positive granules in the proximal straight tubular epithelium became prominent in females on day 50. This paper is the first to describe the development of gender differences in mouse kidney morphology.  相似文献

Although mice kidney morphology shows various sexual dimorphisms, the effect of the estrous cycle has not previously been discussed. In this study, we investigated the effects of the estrous cycle on kidney morphology, including renin-positive areas, of female DBA/2 mice. No effects were confirmed in most of the histometrical parameters, however, the percentage of the renal corpuscles in which cuboidal epithelium covered under 50% of the parietal layer was significantly higher during estrus compared to that during anestrus.  相似文献

To clarify the strain differences in the morphology of the rat kidneys, we investigated the morphometrical characteristics of the kidneys of Slc:Wistar, Slc:SD, and F344/NSlc rats. The diameter of the renal corpuscles in female F344/N rats is smaller than that in female Wistar rats. Although sex differences (males>females) were shown in SD and F344/N rats, no effects of castration were detected in any of the groups. Strain-dependent differences in the percentage of renal corpuscles with a cuboidal parietal layer were found in both male and female groups. The highest percentage of them was noted in male Wistar rats. Effects of castration were observed in female Wistar and male F344/N rats, and the values after castration were significantly higher than those in the intact animals. As for the number of proximal convoluted tubular nuclei, no strain differences were detected in either the male or female groups. Although a sex difference was found in SD rats (female>male), no effects of castration were detected in any of the groups. In female F344/N rats, numerous numbers of PAS-positive granules, which were observed in the proximal convoluted and straight tubular epithelia, were noted. Orchiectomy induced an increase of these granules in male SD and F344/N rats, but ovariectomy showed no effects on them in any strains. This is the first study to clarify the strain differences in the morphological characteristics of the kidneys in ordinary rat strains.  相似文献

In the present study, we histologically and morphometrically investigated species differences in renal structure using laboratory rodents (mice, gerbils, hamsters, rats, and guinea pigs). Morphometric parameters were as follows, 1) diameter of the cortical renal corpuscles, 2) diameter of the juxtamedullary renal corpuscles, 3) percentage of the renal corpuscles with a cuboidal parietal layer, 4) number of nuclei in proximal convoluted tubules (PCTs) per unit area of cortex, 5) semi-quantitative score of the periodic acid-Schiff (PAS) -positive granules in PCTs, and 6) semi-quantitative score of the PAS-positive granules in proximal straight tubules (PSTs). Significant species differences were detected for each parameter, and particularly severe differences were observed in the PAS-positive granules of PCTs and PSTs. Granular scores varied among species and sexes. Vacuolar structures that did not stain with PAS or hematoxylin-eosin were observed in the renal proximal tubules. The appearance and localization of these vacuolar structures differed remarkably between species and sexes.  相似文献

The kidneys of non-diabetic NOD and wild type ICR mice were examined morphometrically at 3 and 6 months of age. Kidney weights and diameter of renal corpuscles of non-diabetic NOD mice were less than those of ICR mice. No lesions were observed in glomeruli or uriniferous tubules. Renin-positive areas were more common in NOD mice than in ICR mice, but no differences were detected in the Western blot analyses.  相似文献

Atrial (A-type) natriuretic peptide (ANP) is vasodilative hormone involved in the regulation of blood pressure and volume homeostasis. In this study, we examined the differences of the auricular and plasma ANP distribution by immunohistochemistry, ultrastructural morphometry, and radioimmunoassay in five strains of mice. The ANP-immunoreactivities of the auricle were most intense in ICR, and moderate in C57BL/6J and C3H/HeN, and weakest in BALB/cA and DBA/2Cr. The number of ANP-granules was greatest in ICR followed by C57BL, C3H or BALB/c, and DBA/2 mice, in this order. The auricular ANP content was highest in ICR, but the plasma ANP concentration was comparable in all strains. The present study demonstrates that there are differences in the ANP circulating system between five strains.  相似文献

The aim of the present study was to investigate histochemically the localization of thyroid-stimulating hormone (TSH) cells and the number of TSH cells in adenohypophysis of ovariectomized adult females, intact females at estrous and diestrous phase of sexual cycles and castrated and non-castrated adult male rats. TSH cells, distributed throughout the pars distalis in all groups, were observed more frequent in areas near to the pars intermedia than other regions. The cells were round, ovoid, stellar-shaped and had contacts with sinusoids. Their nuclei were large and round-shaped. The cells were found mostly single or occasionally in groups of double and triple. The number of TSH cells was higher in estrous than diestrous phase in intact rats. In adenohypophysis of ovariectomized rats, their numbers were less than that of intacts in estrous, but higher than that of found in diestrous phases. Furthermore, the numbers of TSH cells in intact females during estrous phase was higher than those found in intact and castrated male rats. On the other hand, castration reduced the number of TSH cells while ovariectomy had no effect in this respect.  相似文献

Age-related morphological changes were examined in the kidneys of inbred C57BL/6Cr mice maintained in a controlled environment. The specific pathogen free status of animals used in the present study was confirmed by microbiological monitoring. Kidneys were histologically and histometrically investigated at 3, 5, 12, 15, 24 and 27-months-old. Kidney weights did not change with age. Renal corpuscles increased in number at 24- and 27-months-old, but diameter remained constant. The percentage of renal corpuscles with a cuboidal glomerular capsule decreased at 24- and 27-months-old. Score indicating glomerular damage increased from 5- to 27-months-old. Changes to the proximal convoluted tubules were severe. Vacuolar degeneration was observed from 12-months-old. Tubular atrophy was observed at 24- and 27-months-old, with number of nuclei per unit area increasing at the same ages. Amyloidosis and scar lesions were observed at 27-months-old. Focal cell infiltration around vessels was found at 24- and 27-months-old. Electron microscopy at 27-months-old revealed expansion of the mesangial matrices and fusion of foot processes in the glomeruli. Enlarged lysosomes with lipid content were observed in the proximal convoluted tubules.  相似文献

Young intact and adult gonadectomized C3H male and female mice were utilized as bioassay models to detect endocrine disruption chemicals. Animals treated with oestradiol (E) or progesterone (Prog) or E plus Prog were used to assess steroid hormone agonist and antagonist activities of 4-OH-tamoxifen (TAM), diethylstilbestrol (DES) and genistein (GEN) by bioassay. The stimulation or inhibition of mammary growth by TAM depended on sex, the state of animal (intact or gonadectomized), hormonal treatment (Prog, E, E plus Prog) and dose of TAM. TAM stimulated mammary growth in untreated ovariectomized (OV-X) females and in Prog-treated intact males and OV-X females. In intact males, mammary growth was increased by TAM at dose 0.1-1 microg day(-1) and decreased at dose 10 microg day(-1). Mammary growth was inhibited by TAM in Prog-treated intact females and in E or E plus Prog-treated intact and gonadectomized males and females. Uterine weights were increased by TAM in both untreated and treated (E, Prog, E plus Prog) intact and OV-X females; however, seminal vesicle weights were decreased by TAM in both untreated and treated (E, Prog, E plus Prog) intact males. DES alone affected mammary growth (an inverted-U-shaped dose-response curve) both in male and female mice. DES increased uterine weights; however, seminal vesicle weights were decreased. GEN increased mammary and uterine growth in OV-X females, GEN plus Prog stimulated mammary growth in intact males. The results obtained in these studies show clearly that only a bioassay consisting of several endpoints reflective to the mechanism of oestrogen and anti-oestrogen action has the ability to evaluate activities of a molecule.  相似文献

No detailed comparative data are available on the hormonal parameters of horses suffering from a number of diseases. The aim of our study was to measure concentrations of cortisol, adrenocorticotropic hormone (ACTH), serotonin, adrenaline and noradrenaline in horses with various diseases and following surgery, to assess the response of the HPA axis and adrenal medulla. Blood samples were obtained from six groups of horses comprising a total of 119 animals as follows: laminitis, acute abdominal syndrome (AAS), castration surgery, acute diseases, chronic diseases and healthy controls. Serum hormonal concentrations were determined for each group for comparison. Statistically significant differences between all groups and controls were found for cortisol, ACTH (except for castration), serotonin and adrenaline concentrations but only in horses with laminitis and AAS for noradrenaline. No statistically significant differences were found between males and females. The largest changes in the pituitary-adrenal axis activity occurred mainly in acute diseases, laminitis and in the AAS group.  相似文献

1. Plasma concentrations of luteinising hormone (LH), comb size and body weight were measured between 46 and 208 d of age in intact and castrated cockerels from lines of selected and control Australorp domestic chickens. The selected line had been selected for increased rate of lay by reducing oviposition intervals within sequences. The cockerels were reared and maintained on 15.25 h light/d. 2. Concentrations of plasma LH in the intact control cockerels were low in the 'immature' phase (less than 100 d old) and increased during the 'mature' phase (older than 175 d) with a peak occurring when the testes are beginning to grow at the onset of the 'mature' phase. In comparison with the control line of cockerels, the selected line had significantly higher plasma LH concentrations at the onset of the 'mature' phase. Selection had no effect on the mean concentration of plasma LH in either the immature phase or a few weeks after the onset of the mature phase. 3. There was no effect of selection in sibling females for higher rates of lay on changes in comb size or body weight during sexual maturation in the intact cockerels. 4. Castration at 45-46 d of age resulted in increased plasma LH concentrations and no comb growth. Plasma LH concentration increased progressively between 40-208 d. There was no difference between the two lines of cockerels in plasma LH concentration or rate of body growth after castration. 5. It is concluded that selection of females for a change in the rate of egg production has resulted in increased plasma LH concentrations in sibling males around the onset of sexual maturation. Selection appears to have caused this effect by altering an unidentified component of the inhibitory feedback mechanism which controls the tonic secretion of LH.  相似文献

The objective of the present study was to determine if postpubertal boars (12-13 months of age; 156 +/- 8 kg) with large testes had altered hypothalamic control of secretion of luteinizing hormone (LH). Seven boars with the highest estimated 150 d, paired testis weights from a line selected for large testes (769 +/- 60 g = mean weight of excised testes) and 8 boars from a control group (control, 544 +/- 20 g) were tethered in stalls and fitted with indwelling jugular catheters. Males were bled when they were intact, 14 days after castration and during administration of sodium pentobarbital anesthetic (subsequent to castration) to block secretion of endogenous LH-releasing hormone (LHRH). Blood samples were collected at 12-min intervals for 6 hr before and 1 hr after intravenous injection of LHRH in intact and castrated males. During anesthesia, LHRH was administered 4 times at 1-hr intervals and blood samples were collected every 6 min. All samples were analyzed for concentrations of LH and pooled samples were analyzed for concentrations of 17-beta estradiol (E2) and testosterone (T). In intact and castrated males, mean concentrations of LH, frequency and amplitude of pulses of LH, and concentrations of E2 and T were not different between boars of the two groups (P greater than .10). Response to exogenous LHRH was less (P less than .05) in intact males with large testes than in corresponding males from the control group (P less than .05). It is well established that sexual dimorphism exits within the immune system. Females have higher levels of immunoglobulins, greater antibody response to antigens, and higher incidence of autoimmune diseases, such as systemic lupus erythematosus, Grave’s disease, and Hashimoto thyroiditis than males. Spontaneous autoimmune syndromes in mice are more prevalent and of greater severity in females compared with males, and the course of the disease can be modulated by changes in levels of gonadal steroids. A sexual dimorphism is also present in the pituitary-adrenal function: females have higher corticosterone levels and higher corticosteroidogenesis.In the context of the immune-neuroendocrine interactions, we investigated the effects of gonadectomy and sex hormone therapy on endotoxin-stimulated hypothalamo-pituitary-adrenal axis. Whereas endotoxin-induced corticosterone release is invariable throughout the different stages of the oestrus cycle, gonadectomy in both male and female mice leads to enhanced adrenal and immune responses to endotoxin. Interestingly, these enhanced adrenal and immune responses can be completely reversed by testosterone treatment regardless of the sex of the mice. Studies performed over development confirm the role of endogenous testosterone in modulating the endotoxin-induced corticosterone secretion. Indeed, corticosterone response to endotoxin is maximal before puberty when endogenous testosterone levels are low and declines in postpubertal and adult mice.In conclusion, all these data support a sex steroid hormone basis for a neuroendocrine-immunologic sexual dimorphism.  相似文献

An experiment was conducted to determine whether exogenous estradiol-17beta (E2) could restore sexual behavior in castrated rams. The protocol consisted of three sequential 6-wk periods during which rams were studied while 1) intact, 2) bilaterally castrated, or 3) implanted s.c. with two 7.6-cm silastic implants each containing 309+/-16 mg of E2. Rams (classified as female-oriented [FOR, n = 7], male-oriented [MOR, n = 7], or asexual [n = 7]) were subjected to 30-min sexual behavior tests every 2 wk during the ensuing 18 wk. Rams were observed for mounts and ejaculations using two ovariectomized, estrous ewes and two intact males secured in stanchions. Behavioral data were analyzed using the signed rank test, but asexual rams showed no sexual behavior and therefore were not evaluated statistically. Jugular blood was collected prior to castration and at the end of the 18-wk period, and testicular venous (n = 21) and arterial (n = 8) bloods were collected immediately prior to castration. Radioimmunoassay was used to quantify systemic levels of estrone (E1), E2, and testosterone (T) and testicular serum concentrations of oxytocin (OT). Mounting behavior of MOR and FOR declined after castration (P < .05 and P < .10, respectively). Castration reduced the number of ejaculations by FOR (P < .05), but not by MOR (P > .10). Mounting behavior of castrated MOR and FOR was not affected by E2 treatment relative to that observed if castrated only (P > .10). Treatment of asexual rams with E2 did not stimulate sexual behavior in these rams. There were no marked differences (P > .10) among ram groups with regard to serum concentrations of E1, E2, or T prior to castration (overall mean +/- SE, 12.8+/-.7, 7.6+/-.5, and 2,670+/-780 pg/mL, respectively) or any difference (P > .10) in systemic concentration of E1 or E2 among ram groups after rams were implanted with E2 (overall mean +/- SE, 9.7+/-.7 and 9.0 +/-.7 pg/mL, respectively). Serum concentrations of E2 after implantation of the steroid did not differ from those present while rams were intact (P > .10). Testicular venous and arterial serum concentrations of OT were low and did not differ within or between rams. These results suggest that restoration of E2 concentrations to physiological levels in castrated adult rams (regardless of sexual orientation) cannot stimulate or reestablish sexual behaviors to levels observed prior to castration.  相似文献

Seventy-two Angus feedlot bulls were observed to determine the effects of implanting with zeranol (0, 36 or 72 mg; four groups of six/level) at 8 mo of age (d 0), and of castration midway through the feedlot period (d 115) vs remaining intact (six groups/treatment) on sexual, agonistic and handling behaviors. All 36-mg-treated cattle were re-implanted (36 mg) on d 112; 72-mg-treated cattle were not re-implanted. Each group was observed for a total of 8 h at the time of sunset +/- 30 min for sexual and agonistic behaviors during the trial. Observations on handling were made during 15 weighings and on seven occasions in the pens of the animals. Zeranol did not affect sexual or agonistic behavior. Before imposition of castration, implanted bulls entered the scales more quickly, took longer to stand still on the scales and were less quiet than non-implanted bulls (P less than .05). Castration decreased the sexual behaviors of masturbation (.75 vs .08 X pen-1 X h-1; P less than .05), attempted mounts (.75 vs .33 X pen-1 X h-1; P less than .05) and chinning (10.70 vs 4.75 X pen-1 X pen-1; P less than .10) and the agonistic behavior of butting (42.42 vs 31.50 X pen-1 X h-1; P less than .01) in contrast to intact bulls. This study presents the results from a morphometric analysis of 52 dry Retriever dog pelvic bones (30 male, 22 female). A total of 20 parameters were measured using an osteometric board and digital vernier caliper. Six parameters were found to be significantly higher (< 0.05) in males than in females, while one parameter was significantly higher (< 0.05) in females than in males. However, none of the measured parameters demonstrated clear cut‐off values with no intersect between males and females. Therefore, we generated a stepwise discriminant analysis from all 20 parameters in order to develop a possible working equation to discriminate gender from a dog pelvic bone. Stepwise discriminant analysis was used to create a discrimination function: Y = [82.1*PS/AII] ? [50.72*LIS/LI] ? [23.09*OTD/SP] + [7.69*SP/IE] + [6.52*IC/OW] + [7.67*ISA/OW] + [20.77*AII/PS] + [504.71*OW/ISA] ? [90.84*PS/ISA] ? [148.95], which showed an accuracy rate of 86.27%. This is the first study presenting an equation/function for use in discriminating gender from a dog's pelvic measurements. The results can be used in veterinary forensic anthropology and also show that a dog's pelvis presents sexual dimorphisms, as in humans.  相似文献

To determine if cytoplasmic effects have contributed to long-term selection response for increased growth rate in mice, reciprocal cross matings were made between an unselected control line (ICR) and a line (M16) derived from ICR by long-term selection for high postweaning weight gain from 3 to 6 wk of age. Embryos were recovered 2 to 4 d following mating and transferred to pseudopregnant F1 (DBA/2NCrlBR X C57BL/6NCrlBR) females. Thus, all embryos developed in similar uterine and postnatal maternal environments. A total of 122 M16 X ICR and 123 ICR X M16 mice was produced, representing 19 litters from each cross. Litters were standardized at birth to five to seven pups. Litter weights at birth and 1 wk were recorded. Body weights at 2, 3, 4, 5 and 6 wk and weight gain from 3 to 6 wk were obtained. Weights of liver, kidneys, and sc and epididymal fat pads of males were obtained at 6 wk. Females were mated at 8 wk, and litter size at birth was recorded. Least-squares procedures were used to test for differences between reciprocal crosses for all traits. Body weight at 4 wk was higher (P less than .05) for mice with ICR cytoplasm. No other significant differences were detected. There was no evidence that cytoplasmic effects influenced direct or correlated responses to long-term selection for increased postweaning weight gain.  相似文献

Diesel exhaust particles (DEPs) contain polycyclic aromatic hydrocarbons (PAH) that bind to aryl hydrocarbon receptors (AhRs) and decrease sperm production. Since it is not clear if AhR mediates DEP toxicity, we investigated the effect of DEPs in four strains of mice that have different AhR responsiveness. We treated BALB/c, C57BL/6, ICR and DBA/2 mice with DEP suspensions and compared their toxicity in each strain. In both the vehicle- and DEP-treated groups, ethoxyresorufin-O-deethylase (EROD) activity, as an indirect index of AhR activity, was increased in the order of BALB/c > C57BL/6 > ICR > DBA/2. Only BALB/c and C57BL/6 mice had significantly lower daily sperm production (DSP) than vehicle-treated mice. All strains exhibited increased sperm abnormalities. In particular, the C57BL/6, ICR and DBA/2 mice exhibited significantly increased abnormalities. A significant correlation was found between EROD activity and DSP or incidence of morphologically abnormal sperm. These data suggest that DEP toxicity may affect the male reproductive system in an AhR-dependent manner.  相似文献

[目的] 研究结石通片对小鼠的急性毒性作用,对健康大鼠的利尿作用,以及对乙二酰胺致大鼠肾结石的调节作用。[方法] 取ICR小鼠40只,雌、雄各半,随机分为对照组与给药组,每组20只,观察结石通片给药前后小鼠的体重变化和死亡情况;另取SD大鼠36只,雌、雄各半,分为对照组、低剂量组(给予5%结石通片溶液)和高剂量组(给予10%结石通片溶液),统计各组大鼠24 h排尿量;并采用42只雄性SD大鼠,考查结石通片对给予乙二酰胺(2%掺喂)后大鼠体重、肾重和肾重比的影响。[结果] 给予结石通片溶液后,连续观察7 d,小鼠的各项生理活动未见明显异常,给药组与对照组相比体重差异不显著(P>0.05);与对照组比较,给予结石通片溶液后大鼠24 h内排尿量明显增加(P<0.01);与对照组比较,给药组随着给药剂量的增加,结晶和结石的数量极显著(P<0.01或P<0.001)减少,形成结石的动物数量极显著(P<0.01或P<0.001)减少。[结论] 结石通片对小鼠无明显急性毒性作用,对大鼠利尿作用明显,并对乙二酰胺致大鼠肾结石具有明显抑制作用。  相似文献
