氟喹诺酮类药物的多残留酶免疫分析研究

摘    要:以制备的多抗血清为基础,建立了氧氟沙星(OFL)、二氟沙星(DIF)和诺氟沙星(NOR)的多残留间接竞争酶免疫吸附测定法(Ci ELISA法)。OFL、 DIF和NOR的标准曲线的线性回归方程分别为y=-0.1984x+0.9687 (r=0.9885)、y=-0.1738x+0.9150 (r=0.9985)、y=-0.2004x+0.9572(r=0.9954);中值(IC50)分别为231.57、245.32 、186.14 ng/ml ;最低检测限(LOD)分别为1.62、1.86、1.07 ng/ml;标准曲线在2~10000 ng/ml之间均有良好的线性关系(r≥0.9885)。方法的批内CV≤5.44%,批间CV≤12.45%。当OFL、DIF或NOR以浓度15~10000 ng/ml在牛奶中添加时,药物能较好地被回收,回收率均在71.70%~94.53%之间。

Abstract:The antiserum was prepared with the mixture of OFL-BSA,DIF-BSA and NOR-BSA.The Ci-ELISA method for the detection of ofloxacin,difloxacin and norfloxacin was established with it.To ofloxacin,difloxacin and norfloxacin.The regression equations about their standard curves were y=-0.1984x 0.9687(r=0.9885),y=-0.1738x 0.9150(r=0.9985) and y=-0.2004x 0.9572(r=0.9954),respectively.The 50% inhibition concentration(IC50) were 231.57,245.32 and 186.14ng/ml,respectively.The limits of detect(LOD) were 1.62,1.86 and 1.07ng/ml,respectively.The coefficients of variation for the Ci-ELISA method were less than 5.44% in intra-assay and 12.45% in inter-assay.The recoveries in milk ranged from 71.70% to 94.53%.
