基因枪法转基因水稻后代农艺性状的表现

摘    要: 通过基因枪转化法,将潮霉素磷酸转移酶基因(hpt)导入粳稻品种77170和中花9号,获得转化再生植株,选其中7个结实较好的株系为试验材料,对其自交后代(T1 和T2)的株高、穗数、穗长、穗粒数、结实率和千粒重等6个农艺性状进行调查分析。结果显示,转基因植株后代6个农艺性状都不同程度地发生了变异,SG-1的千粒重变异较大,SG-2穗数变异较大,SG-3-1结实率变异较大,SG-3-2在6个性状上都有较大的变异,SG-15主要是在株高、穗长、穗粒数以及穗数和千粒重5个性状上有较大变异。产生这些变异的主要原因是在组织培养过程中发生了体细胞无性系变异,导入外源基因并没有直接影响农艺性状的表达。在T1和T2代中,千粒重和结实率的表现基本一致,而株高、穗数、穗长和穗粒数的遗传表现还不够稳定。因此,在T1代选育时,应注重选择千粒重和结实率表现好的株系,而对本研究涉及的其它性状可不作严格选择。

Abstract:Japonica rice cv 77-170 and ZH9 were transformed with pBI222 carrying hygromycin phos- photransferase gene(hpt) by using biolistic bombardment method, and regenerated plants were obtained. Seven seed-setting regenerants(SG-1, SG-2, SG-3-1, SG-3-2, SG-15 was derived from 77-170, and YW2- I-6, YW,-5-8 was derived from ZH9) were selected as materials to analyse the variation of 6 agronomic traits(plant height, number of panicle, panicle length, number of spikelets per panicle, seed-setting rate and grain weight ) in their T1 and T2 generation. Different variation of 6 agronomic traits were observed in the progenies of different transgenic rice plants. These changes mainly due to the somaclonal variation, and introduction of hPt did not directly affect the performance of agronomic traits in the progeny of trans- genic plants. The variation at seed-setting rate and grain weight were basically similar between T1 and T2 generation,so the plant lines which performed better on the two traits should be selected in T1 generation, and the traits of plant height, number of panicle, panicle length and number of spikelets per panicle may not be selected in early generation for their unstable inheritance from T, generation to T2.
