中国主栽银耳配对香灰菌的系统发育和遗传多样性

摘    要:【目的】了解中国主栽银耳配对香灰菌的分类地位及遗传多样性,为香灰菌资源利用和育种提供分子生物学依据。【方法】采用18S-28S rDNA间隔区(ITS4/ITS5)的PCR扩增和内部简单重复序列多态性(ISSR)分子标记,对中国主栽银耳配对香灰菌的遗传多样性进行分析。【结果】16株供试菌株ITS序列分析表明,供试菌株与Annulohypoxylon stygium关系最为接近,同源性高达99%。从70个ISSR引物中筛选出了11个多态性好的引物,共获得62条ISSR标记。以相关系数0.7为阈值,将16株菌划为5个遗传组,菌株间遗传差异较大,遗传多样性较高。【结论】中国主栽银耳地区的香灰菌遗传资源较为丰富,这为利用香灰菌资源选育新品种提供了可能。
