喀斯特地区野生旱半夏组培快繁技术

摘    要:将野生旱半夏置于35 ℃的高温条件下处理21 d后剥取茎尖分生组织,采用植物组织培养技术培育野生旱半夏脱毒试管苗。通过对培养基添加不同种类、不同浓度的外源激素,对诱导、增殖和生根系列培养基优化选择进行研究。试验结果表明,野生旱半夏茎尖分生组织培育试管苗的最佳诱导培养基组合为MS+6BA 2.0 mgL+NAA 0.3 mgL,增殖培养基组合为MS+6BA 2.0 mgL+NAA 0.2 mgL,生根培养基组合为MS+NAA 0.15 mgL。

Abstract:Shoot apex meristem of wild drought Pinellia ternate was stripped after treatment at high temperature at 35 ℃ for 21 days.Plant tissue culture technology was adopted to cultivate virus free test tube plantlets of wild drought Pinellia ternate.Optimized selection of culture medium for induce,proliferation and rooting series was researched by adding different kinds and different concentrations of exogenous hormones to culture medium.Test results showed that best inducing medium combination of cultivation test tube seedlings of shoot apex meristem for wild drought Pinellia ternate was MS+6BA 2.0 mgL+NAA 0.3 mgL,proliferation medium combination was MS+6BA 2.0 mgL+NAA 0.2 mgL,rooting medium combination was MS+NAA 0.15 mgL.
