孕早期家兔子宫内膜细胞的分离培养与形态观察

摘要:
为探讨子宫内膜细胞的分离与体外培养条件、生长特点及其增殖过程,采用不同消化方法进行了孕早期家兔子宫内膜细胞的消化和分离培养试验。结果表明,多数情况下采用胰蛋白酶消化获得的细胞数量少且活率低于60%,而采用1 g/L胶原酶 型消化获得的细胞数量多达1×106/cm3,且活率大于90%。经74 μm(200目)滤网过滤分离培养出多角形和梭形2种形态的基质细胞及铺路石状的腺上皮细胞,并均能在体外培养和传代。

Isolation,culture and morphological observation of early pregnant rabbit
Abstract:
This paper focuses on optimal condition of the isolation and in vitro culture of endometrium cells of early pregnant rabbit,which were also dealt with the morphological modality growth and proliferation characteristics.Different digestion methods and isolation methods used to prepare the cells were estimated by the cell culture.The results show that in most conditions,the number of cells was small and the viability ratio was lower than 60% using trypsin to digest cells,while using the density of 1 g/L to digest cells,the number of cells was up to 1×106/cm3 and the viability ratio was 90%.The stromal cell and the epithelial cell were able to be separated through 74 μm filter.Two kinds of the cells could be cultured to a full monolayer and regenerated in vitro.
