利用PCR技术从雄性中国荷斯坦牛的基因组DNA中克隆了Y染色体性别决定基因(Sex-determining Region on the Y Chromosome,SRY)的编码区全长序列,构建了表达载体pET-28a/SRY,并将其在大肠杆菌(E.coli)中进行了诱导表达,对表达产物进行了检测。结果表明,SRY基因编码区长687 bp,编码229个氨基酸;表达载体pET-28a/SRY构建成功;表达产物中含有相对分子质量为33ku的SRY蛋白。 |

Cloning of Holstein cow's SRY ORF and its expression in E.coli |

Abstract: |
The gene of SRY ORF was cloned using Holstein cow genome.The total coding region is 687 bp,encoding a peptide with 229 amino acid residues.The Holstein cow’s SRY ORF was cloned to EcoRⅠ and SalⅠ sites of pET-28a(+) to construct an expression plasmid pET-28a/SRY.The expression plasmid was transformed to E.coli BL21 (DE3),induced by IPTG.SDS-PAGE analysis and confirmed that the Holstein cow’s SRY peptide was highly expressed at 4 h induction at 37 ℃.The SRY protein was expressed from totle protein of E.coli BL21 (DE3). |
